摘 要:【目的】探究中草药黑老虎(Kadsura coccinea)醇提物对尖吻蝮(Deinagkistrodon acutus)蛇毒中主要酶类的抑制作用。【方法】用体积分数为75%的乙醇溶液浸泡黑老虎后再进行超声波处理得到醇提物;通过体外酶活抑制实验分别检测醇提物对尖吻蝮蛇毒中磷脂酶A2、蛋白水解酶、透明质酸酶、类凝血酶的抑制作用。【结果】黑老虎醇提物对尖吻蝮蛇毒中的蛋白水解酶、磷脂酶A2和透明质酸酶具有明显的抑制作用。当蛇毒与醇提物的质量比分别为1∶100,1∶10和1∶5时,后者能抑制蛇毒中98%磷脂酶A2活性、79%蛋白水解酶活性和65.14%透明质酸酶活性。同时,醇提物能够明显抑制尖吻蝮蛇毒水解纤维蛋白原和类凝血酶的活性。【结论】研究结果证实了黑老虎醇提物能有效抑制尖吻蝮蛇毒中的主要酶类,为深入研究黑老虎药材的抗蛇毒机制提供了理论依据。[Purposes]To investigate the inhibitory effect of the Chinese herbal medicine Kadsura coccinea ethanol extract on the main enzymes of Deinagkistrodon acutus venom.[Methods]Kadsura coccinea ethanol extract was obtained by immersing Kadsura coccineain 75% ethanol solution and then ultrasonic treatment.The inhibitory effect of the ethanol extract of K.coccinea on the phospholipase A2,proteolytic enzyme,hyaluronidase and thrombin-like enzymeof D.acutus venom was detected in vitro by enzyme activity inhibition experiments.[Findings]K.coccinea ethanol extract have significant inhibitory effects on proteolytic enzymes,phospholipase A2 and hyaluronidase in D.acutus venom.When the mass ratio of snake venom to the alcohol extract was 1∶100,1∶10 and 1∶5 respectively,the latter could inhibit 98% phospholipase A2 activity,79% proteolytic enzyme activity and 65.14% hyaluronidase activity in snake venom.At the same time,the ethanol extract can significantly inhibit the hydrolyzed fibrinogen activity and thrombin activity of D.acutus venom.[Conclusions]It confirmed that the ethanol extract of the Chinese herbal medicine K.coccinea can effectively inhibit the main enzymes in D.acutus venom,which provides an theoretical basis for further research on the mechanism of anti-venom of this medicine.
