摘 要:目的:利用网络药理学探究马齿苋活性成分缓解结肠炎的作用机制。方法:基于TCMSP数据库获得其活性成分和对应靶标,利用STRING进行靶标间互作分析并筛选核心靶标,经KEGG富集分析得到马齿苋治疗疾病的相关信号通路。基于Drugbank数据库获取结肠炎的相关疾病靶标,与马齿苋成分靶标进行映射,得到马齿苋治疗结肠炎的作用机制。结果:共筛选获得马齿苋活性成分花生四烯酸、β-谷甾醇、山柰酚、木犀草素、异甜菜素、槲皮素等10个,其对应靶标共187个,核心靶标共62个,经KEGG富集分析发现核心靶标与癌症、病毒感染、炎症性肠炎和免疫调节相关通路密切相关,并通过ABCC4、ABCG2、ACAT1、ALB、ALOX5、CYP3A4、IFNG、IKKA、MPO、NOS2、NQO1、NR1I2、PPARG、PTGS1、PTGS2、TNF、XDH起到治疗结肠炎的作用。结论:马齿苋可通过抗炎、抗氧化和调节细胞增殖与凋亡发挥多靶点、多通路缓解结肠炎的作用。Objective:To explore the mechanism of the active ingredients of Herba Portulacae in relieving colitis based on network pharmacology.Methods:The active ingredients and corresponding targets were obtained based on the TCMSP database,then STRING database was used to analyze the interaction among the targets and to screen the core target,which was analyzed by KEGG enrichment to obtain the signal pathway related to Herba Portulacae.Colitis-related disease targets were obtained through the Drugbank database and the targets of the ingredients of Herba Portulacae were mapped to obtain the mechanism of treating colitis by Herba Portulacae.Results:A total of 10 active ingredients such as arachidonic acid,β-sitosterol,kaempferol,luteolin,isobetain,quercetin were obtained,with 187 corresponding targets and 62 core targets.By KEGG enrichment analysis,it was found that the core targets were closely related to cancer,viral infection,inflammatory enteritis and immune regulation-related pathways,and they played a role in the treatment of colitis through targets ABCC4,ABCG2,ACAT1,ALB,ALOX5,CYP3A4,IFNG,IKKA,MPO,NOS2,NQO1,NR1I2,PPARG,PTGS1,PTGS2,TNF and XDH.Conclusion:Herba Portulacae can play a multi-target and multi-channel role in alleviating colitis through anti-inflammatory,antioxidant,and regulating cell proliferation and apoptosis.
