摘 要:目的建立八色荧光38个基因座的复合扩增体系。方法采用八色荧光标记技术,对38个基因座Amelogenin、D5S818、D21S11、D7S820、CSF1PO、D2S1338、D3S1358、vWA、D8S1179、D16S539、D1S1677、PentaD、TPOX、TH01、D22S1045、D18S51、FGA、D6S1043、D13S317、D12S391、PentaE、D2S441、D1S1656、D19S433、D10S1248、D11S4463、D1S1627、D3S4529、D1GATA113、D17S974、D6S1017、D4S2408、D9S2157、D6S474、D2S1776、D18S853、D20S482及D14S1434进行复合扩增,应用GA118-24B遗传分析仪进行电泳检测,应用Gene Typer V1.0软件进行数据分析。结果本文成功建立了38个基因座的八色荧光复合扩增体系,基因座间均衡性良好,阳性对照分型结果准确。结论包含38个基因座的八色荧光复合扩增体系构建成功,为后续更多八色试剂盒的开发奠定了基础。在提高我国DNA检验试剂水平、遗传分析仪检测能力和数据分析软件研究方面具有重要意义。Objective To construct a multiplex amplification system of 38 loci with eight-dye fluorescence. Methods Eight-dye fluorescent labeling technology was used to multiplex 38 loci(Amelogenin, D5 S818, D21 S11, D7 S820, CSF1 PO, D2 S1338, D3 S1358, vWA, D8 S1179, D16 S539, D1 S1677, PentaD, TPOX, TH01, D22 S1045, D18 S51, FGA, D6 S1043, D13 S317, D12 S391, PentaE, D2 S441, D1 S1656, D1 S1656, D19 S433, D10 S1248, D11 S4463, D1 S1627, D3 S4529, D1 GATA113, D17 S974, D6 S1017, D4 S2408, D9 S2157, D6 S474, D2 S1776, D18 S853, D20 S482 and D14 S1434). GA118-24 B genetic analyzer was used for electrophoresis detection, and Gene Typer V1.0 software was used for data analysis. Results The eight-dye fluorescence multiplex amplification system of 38 loci was successfully established, and it was accurate and stable. Conclusion The platform of eight-dye fluorescence detection and multiplex amplification system of 38 loci was constructed successfully, which laid a foundation for the development of more eight-dye kits. It plays an important role in improving the level of DNA testing reagents, the detection ability of genetic analyzer and the research of data analysis software.
