摘  要:目的建立20个STR、36个Y-STR、3个Y-indel及Amelogenin基因座的复合扩增体系,并对其性能指标进行验证。方法采用八色荧光标记技术,选择涵盖常染体和Y染色体的60个基因座,合成引物,进行复合扩增以及毛细血管电泳检测;并对其准确性、灵敏度、种属特异性、混合样本、稳定性及检材适应性等系统性能指标进行验证。结果采用本文体系分型结果准确稳定,检测灵敏度达0.125ng/μL,能够满足对混合检材以及建库过程中批量检测的要求,具备良好的稳定性和种属特异性。结论本文建立的60个基因座的八色荧光染料标记的STR及Y-STR复合扩增体系分型准确稳定,在今后法庭科学以及数据库建设等方面具有广阔的应用前景。Objective To deveplope a new multiplex PCR system comprised of 20 STR, 36 Y-STR, 3 Y-indel and Amelogenin markers and varify its performance index. Methods Using eight color fluorescent labeling technique, 60 locus characterized were amplified simultaneously in a multiplex PCR system, PCR product was analyzed in a ABI 3500 XL Genetic Analyzer. The system performance indexes, such as accuracy, specificity, sensitivity and stability of the system and its validation on the mixtures were verified. Results The resulsts of this system were stable, accurate, and sensitive. The detection sensitivity was 0.125 ng/μL, which could meet the requirements of mixed materials and batch detection in the process of data construction, and had good stability and specificity. Conclusion The multiplex PCR system is effective and reliable for forensic routine DNA analysis. It is very helpful for constructing Chinese forensic database and population genetic research.
