摘 要:本研究以棉花根际土壤为试材,采用土壤稀释平板法和平板对峙法筛选棉花黄萎病的高效拮抗菌,并结合形态观察和分子鉴定方法对拮抗菌株进行鉴定。结果表明,筛选的8株菌对棉花黄萎病菌(大丽轮枝菌)均有一定的拮抗作用,其中,B51菌株的拮抗效果最好,抑制率为42.86%。分子鉴定结果显示,基于rDNA-ITS、EF1α、β-tubulin和ACT基因扩增测序后,经GenBank比对,B11、B51、C31和C91菌株与粉红粘帚霉Clonostachys rosea相似度为97%~98%。基于rDNA-ITS基因比对,C41、A21和C421菌株与高山被孢霉Mortierella alpina的相似度为99%,C11菌株与球毛壳菌Chaetomium globosum相似度为100%。In this study, cotton rhizosphere soil was used as research material to screen high-efficient antagonistic bacteria against Verticillium wilt by soil dilution plate method and plate confrontation method, and the antagonistic strains were identified by morphological observation and molecular identification methods. The results showed that all the 8 strains had a certain antagonistic effect against Verticillium dahliae, and B51 strain had the best antagonistic effect against V. dahliae, the inhibition rate was up to 42.86%.The results of molecular identification showed that the homologies of the strains B11, B51, C31 and C91 were ranging from 97%-98%with Clonostachys rosea based on rDNA-ITS, EF1α, β-Tubulin and ACT genes. The similarity of the strains C41, A21 和 C421 was up to 99% with Mortierella alpina;and the similarity between strain C11 and Chaetomium globosum was 100%.
