摘  要:为筛选海岛棉最优的愈伤组织诱导条件,以3个新疆海岛棉品种无菌苗下胚轴为外植体诱导愈伤组织,设置不同培养温度和光照组合,研究海岛棉出愈率与组织形态特征。结果表明:全光照是海岛棉形成愈伤组织最适宜的诱导条件,黑暗条件下新海40、新海44未能形成愈伤组织;相比于25℃,28℃更适合海岛棉愈伤组织的诱导;3个海岛棉品种愈伤组织颜色以黄绿色、绿色为主,质地较疏松。本研究结果可为海岛棉愈伤组织诱导、植株再生及优化提供借鉴和参考。In order to select the optimal conditions for callus induction of Gossypium barbadense,hypocotyls of sterile seedlings of three island cotton varieties in Xinjiang were used as explants to induce callus,and different combinations of culture temperature and light were set up to study the callus induction rate and tissue morphological characteristics of island cotton.The results showed that:Full light was the most suitable induction condition for callus formation of island cotton,but Xinhai 40 and Xinhai 44 failed to form callus under dark conditions;Compared with 25℃,28℃was more suitable for callus induction;The callus of 3 island cotton varieties were mainly yellow-green or green with loose texture.The results of the study could provide references for callus induction,plant regeneration and optimization of island cotton.
