摘  要:目的观察卵蛋白诱导哮喘过程中尾静脉注射人脐带间质干细胞来源的外泌体(huc MSC-ex)的小鼠肺部及全身炎症反应。方法在第0、7天腹腔注射10%卵蛋白和氢氧化铝混合液1 m L致敏小鼠。1周后将小鼠随机分为huc MSC-ex组、哮喘组、对照组。huc MSC-ex组雾化吸入1%卵蛋白10 m L,每天1次,每次30 min,持续2周,并于每次雾化前尾静脉注射200μg/m L的huc MSC-ex。哮喘组尾静脉注射同体积的PBS后,雾化吸入1%卵蛋白10 m L,每天1次,每次30 min,持续2周。对照组雾化吸入10 m L PBS,持续2周。末次激发48 h后,分别取肺组织、肺泡灌洗液(BALF)及血清。HE染色观察肺组织病理结构。瑞氏染色观测BALF中炎症细胞的数量及比例。ELISA检测小鼠血清中Th2细胞样炎症因子IL-4、IL-5、IL-13。结果与对照组相比,哮喘组肺组织气道壁增厚,管腔不规则,气道周围水肿及炎症细胞浸润明显增加;huc MSC-ex组气道壁无明显增厚,管腔相对规则,气道周围水肿及炎症细胞浸润减轻。与对照组比较,哮喘组BALF中白细胞总数以及嗜酸性粒细胞和中性粒细胞的比例升高(P均<0.05);与哮喘组比较,huc MSC-ex组BALF中白细胞总数以及嗜酸性粒细胞和中性粒细胞的比例降低(P均<0.05)。与对照组比较,哮喘组血清炎症因子IL-4、IL-5、IL-13水平升高(P均<0.05);与哮喘组比较,huc MSC-ex组血清炎症因子IL-4、IL-5、IL-13水平明显下降(P均<0.05)。结论卵蛋白诱导哮喘过程中尾静脉注射huc MSC-ex的小鼠肺部及全身炎症反应减轻。Objective To investigate the effects of exosomes derived from human umbilical cord mesenchymal stem cells(huc MSC-ex)on asthmatic mice induced by OVA.Methods The mice were sensitized with 1 m L intraperitoneal administration of 10%OVA plus aluminum hydroxide on day 0 and 7.The mice were then divided into the control group,asthma group,and huc MSC-ex treatment group after one week.In the huc MSC-ex group,the mice were challenged by 1%OVA 10 m L once a day for half an hour for 2 weeks,and 200μg/m L huc MSC-ex was injected into the tail vein before each atomization.The mice in the asthma group were injected with PBS as the same volume as exosomes,and then 1%OVA 10 m L was given once a day for half an hour for 2 weeks.The mice in the control group were given inhalation of PBS and 1%OVA 10 m L once a day for half an hour for 2 weeks.Lung tissues,alveolar lavage fluid(BALF)and serum were obtained at 48 h after the last stimulation.The number and proportion of inflammatory cells in the BALF was measured by Wright’s staining and cell count.The Th2-like inflammatory cytokines IL-4,IL-5,and IL-13 in the serum of mice were detected by ELISA.Results Compared with the control group,the airway wall was thickened,the lumen was irregular,the peri airway edema and inflammatory cell infiltration were significantly increased in the asthma group.Conversely,it showed no significant thickening of the airway wall,and the lumen was relatively regular,and the peri airway edema and inflammatory cell infiltration were reduced in the huc MSC-ex intervention group.Compared with the control group,the number of white blood cells and ratio of eosinophils and neutrophils in BALF increased in the asthma group(all P<0.05).In contrast,huc MSC-ex reduced the number of white blood cells and ratio of eosinophils and neutrophils in BALF compared with those of the asthma group(all P<0.05).In addition,the serum levels of inflammatory cytokines IL-4,IL-5 and IL-13 increased in the asthma group as compared with those of the control group(all P<0.05).Com
