摘 要:目的基于多聚(ADP-核糖)聚合酶-1(PARP-1)/p53信号通路探讨金莲花总黄酮对人乳腺癌MCF-7细胞增殖的影响。方法将人乳腺癌MCF-7细胞分为低、中、高剂量实验组、对照组,分别予以100,200,300μg·mL-1金莲花总黄酮和等量生理盐水。用CCK-8法测定人乳腺癌MCF-7细胞增殖抑制率;用平板细胞克隆形成实验观察人乳腺癌MCF-7细胞克隆形成情况;用蛋白免疫印迹法检测人乳腺癌MCF-7细胞PARP-1、p53蛋白的表达水平。结果干预48 h时,低、中、高剂量实验组人乳腺癌MCF-7细胞的增殖抑制率分别为(30.65±3.39)%,(44.28±3.76)%和(59.22±4.03)%;对照组、低、中、高剂量实验组克隆形成率分别为(84.52±3.11)%,(49.74±2.86)%,(37.21±3.59)%和(10.32±2.64)%;PARP-1蛋白相对表达量分别为0.92±0.08,0.85±0.05,0.28±0.12和0.09±0.06;p53蛋白相对表达量分别为0.93±0.07,0.73±0.11,0.49±0.10和0.26±0.08。上述指标,对照组分别与低、中、高剂量实验组比较,差异均有统计学意义(均P<0.05)。结论金莲花总黄酮可显著抑制人乳腺癌MCF-7细胞增殖,促进其凋亡,其作用机制可能与抑制PARP-1/p53通路的信号转导相关。Objective Based on the poly(ADP-ribose)polymerase-1(PARP-1)/p53 signaling pathway to explore the effects of total flavonoids of nasturtium on the proliferation of human breast cancer MCF-7 cells. Methods Human breast cancer MCF-7 cells were divided into the exp-L/-M/-H groups and the control group, and were given 100, 200, 300 μg·mL-1 total flavonoids of nasturtium and the same amount of saline respectively. The CCK-8 method was used to determine the proliferation inhibition rate of human breast cancer MCF-7 cells;The clone formation of human breast cancer MCF-7 cells was observed by plate cell clone formation experiments;Western blotting method was used to detect the expression levels of PARP-1 and p53 proteins in human breast cancer MCF-7 cells. Results After 48 h of intervention, the proliferation inhibition rates of human breast cancer MCF-7 cells in the exp-L/-M/-H groups were(30.65±3.39)%,(44.28±3.76)%, and(59.22±4.03)%;the colony formation rates in the control group and the exp-L/-M/-H groups were (84. 52 ± 3. 11) %,(49. 74 ± 2. 86) %(37. 21 ± 3. 59) %,and(10. 32 ± 2. 64) %;The relative expression levels of PARP-1 protein in human breast cancer MCF-7 cells were 0. 92 ± 0. 08,0. 85 ± 0. 05,0. 28 ± 0. 12,0. 09 ± 0. 06;the relative expression levels of p53 protein were 0. 93 ± 0. 07,0. 73 ± 0. 11,0. 49 ± 0. 10,and 0. 26 ± 0. 08. For the above indicators,the control group was compared with the exp-L/-M/-H groups,and the differences were statistically significant(all P < 0. 05). Conclusion Total flavonoids of nasturtium could significantly inhibit the proliferation and promote apoptosis of human breast cancer MCF-7 cells,and its mechanism may be related to the inhibition of signal transduction of PARP-1/p53 pathway.
