摘  要:目的探究山奈酚对慢性应激抑郁模型老年大鼠抑郁样行为的影响。方法用42 d不同应激源暴露刺激的方法诱导建立慢性应激抑郁大鼠模型,将建立的40只慢性应激抑郁大鼠随机分为模型组(n=20)与实验组(n=20),同时选择20只正常大鼠作为对照组。实验组大鼠灌胃50 mg·kg-1山奈酚,对照组与模型组灌胃等量生理盐水,每日1次,连续干预4周。用糖水偏好实验评价各组大鼠的抑郁样行为变化;用蛋白免疫印迹法检测海马组织脑源性神经营养因子(BDNF)、神经生长因子(NGF)蛋白表达水平。结果对照组、模型组与实验组大鼠糖水偏好度分别为(92.03±7.21)%,(34.69±6.52)%,(70.94±8.36)%;海马组织BDNF蛋白相对表达量分别为0.93±0.07,0.14±0.09,0.59±0.14;NGF蛋白相对表达量分别为0.98±0.02,0.31±0.11,0.74±0.12。上述指标,模型组分别与假手术组和实验组比较,差异均有统计学意义(均P<0.05)。结论山奈酚可改善慢性应激抑郁模型老年大鼠的抑郁样行为。Objective To explore the effect of kaempferol on depression-like behavior in elderly rats with chronic stress depression. Methods 42 d of exposure to different stressors were used to induce the establishment of the chronic stress depression rat models, 40 successfully established chronic stress depression rats were randomly divided into the model group(n= 20) and the test group(n= 20), and 20 normal rats were selected as the control group. Rats in the test group were gavaged with kaempferol(50 mg·kg-1·d-1), and the control group and the model group were gavaged with the same amount of saline, once daily and for 4 weeks. The sugar-water preference experiment was used to evaluate the depression-like behavior changes of rats in each group;The protein expression levels of brain-derived neurotrophic factor(BDNF) and nerve growth factor(NGF) in hippocampus were detected by Western blot. Results The sugar water preference of the control group, the model group and the test group were(92.03±7.21)%,(34.69±6.52)%,(70.94±8.36)%;The relative expression levels of BDNF protein in hippocampus were 0.93±0.07, 0.14±0.09, 0.59±0.14;The relative expression levels of NGF protein were 0. 98 ± 0. 02,0. 31 ± 0. 11,0. 74 ± 0. 12;There were statistically significant differences in the above indexes between the model group and the control group/the test group( all P < 0. 05). Conclusion Kaempferol can improve the depression-like behavior of chronic stress depression model elderly rats.
