摘  要:为了拓展鱼胶原的应用范围,研究了采用琥珀酸酐酰化改性制备水溶性鱼胶原,并结合氨基酰化率测定、凝胶电泳、红外光谱、差示扫描量热、表面张力分析、等电点和溶解性测试等手段考察了改性对鱼胶原结构性质的影响。结果表明:当琥珀酸酐的用量为0.2(质量比)时,胶原的氨基酰化率达到约80%,之后酰化率增长缓慢;随着胶原酰化程度的提高,改性胶原的相对分子质量增加,但其热稳定性和等电点逐渐降低;改性前后胶原的红外光谱与表面活性都没有明显变化;当酰化率大于70%时,改性鱼胶原的等电点约为4,在中性条件下具有良好的水溶性。In order to expand the application range of fish collagen,water-soluble fish collagen was prepared by succinic anhydride acylation and the effects of modification on the structure and properties of fish collagen were investigated by aminoacylation rate determination,gel electrophoresis,infrared spectroscopy,differential scanning calorimetry,surface tension analysis,and isoelectric point and solubility tests.The results show that when the dosage of succinic anhydride is 0.2(w/w),the amino acylation rate of collagen reaches about 80%,and then the acylation rate increases slowly.With the increase of the degree of acylation of collagen,the molecular weight of succinylated collagen increases,while its thermal stability and isoelectric point gradually decreases.The infrared spectra and surface activity of the succinylated collagen does not change significantly.When the acylation rate is more than 70%,the isoelectric point of the succinylated fish collagen is about 4,and it has good water solubility under neutral conditions.
