摘  要:目的分析T2DM患者外周血环状RNA(circRNA)差异表达谱,探讨circRNA与T2DM的关系。方法选取2019年9~10月于常州市第二人民医院内分泌科就诊的新诊断T2DM患者20例(T2DM组),同期体检健康人群20名(NC组)。采用表达谱芯片检测两组外周血样本,筛选circRNA差异谱后扩大样本量进行q RT-PCR验证,并预测与circRNA结合的靶miRNA和相关信号通路。结果 circRNA 5139个上调,5422个下调,19个显著改变[P<0.05,差异倍数(FC)>1.5]。取FC改变最明显的5个circRNA进行qRT-PCR验证发现,hsacirc401724、hsacirc100476表达分别上调2.37、1.43倍(P<0.05),hsacirc100241、hsacirc074535、hsacirc028152表达无明显改变(P>0.05)。通过基因软件预测miR-7160-5p、miR-3671、miR-4659a-3p、miR-93和miR-4635是hsacirc401724最高匹配值结合位点;PI3K-Akt是hsacirc401724参与最多的信号通路。结论T2DM患者hsacirc401724表达显著上调,可能通过靶miRNAs调控相关信号通路参与T2DM发生发展。Objective To investigate the differential expression profile of peripheral blood circRNA in type 2 diabetes mellitus(T2DM),and to explore the relationship between circRNA and the development Methods A total of 20 patients with newly diagnosed T2DM(T2DM group)were enrolled in this study form Changzhou Second People’s Hospital,and meanwhile,20 healthy subjects were enrolled as control(NC)group.Peripheral blood samples of the two groups were detected by circRNA microarray.After screening differentially expressed circRNAs,the sample size was expanded for q RT-PCR validation,and target miRNAs bound to circRNA and related signaling pathways were predicted.5139 circRNAas were up-regulated while 5422 were down-regulated.Among them,there were 19 circRNAs showed significant changes(P<0.05,FC>1.5).The 5 circRNAs with the largest FC changes were selected for qRT-PCR verification,and the expression levels of hsacirc401724 and hsacirc100476 were up-regulated by 2.37 and 1.43 times(P<0.05).There was no significant change in the expression levels of hsacirc10024,hsacirc074535 nor hsacirc028152(P>0.05).Bioinformatics techniques showed that miR-7160-5 p,miR-3671,miR-4659 a-3 p,miR-93 and miR-4635 were the highest matched binding miRNAs for hsacirc401724.PI3 k-Akt is the signaling pathway with the most involved hsacirc401724 associated Conclusion Hsacirc401724 was significantly up-regulated in T2DM patients,and may be involved in the occurrence and development of diabetes through the regulation of related signaling pathways by target miRNAs.
