摘 要:以热带高抗大斑病玉米种质CML493幼苗为试验材料,采用人工接种大斑病病菌处理,采用甲基化敏感扩增多态性(MSAP)技术和转录组测序(mRNA-seq)技术,对经过接种大斑病病菌处理不同时间的CML493基因组DNA胞嘧啶甲基化进行DNA甲基化模式、DNA甲基化水平分析和mRNA表达分析。结果表明,接种大斑病病菌6、12 d的甲基化敏感扩增多态性分别为78.21%和76.13%,甲基化敏感扩增单态性分别为21.79%和23.87%。接种大斑病病菌6 d时,总甲基化率上升5.49%,全甲基化率上升14.24%,半甲基化率上升6.11%。接种大斑病病菌12 d时,总甲基化率上升6.40%,全甲基化率上升16.73%,半甲基化率上升5.57%。接种大斑病病菌0、6、12 d共检测到2298个共表达基因,共检测到1434个共表达上调基因,检测到769个共表达下调基因,上调基因数目显著多于下调基因数目。Tropical maize germplasm CML493 were artificial inocuting setosphaeria turcica in this study.The leaves surface present bit spot then gradually increase and enlargement along the extension of time.Methylationsensitive amplification polymorphism(MSAP)technique and transcriptome sequencing(mRNA-seq)technique were used to analyze the DNA methylation pattern,DNA methylation level and mRNA expression of CML493.The results showed that the methylation sensitive amplification polymorphisms of each treatment group were 78.21%and76.13%,and the methylation sensitive amplification polymorphisms of each treatment group were 21.79%and23.87%.The total methylation rate increased by 5.49%,the total methylation rate increased by 14.24%,and the semi-methylation rate increased by 6.11%.When inoculated with bacteria solution after 6 days,the total methylation rate increased by 6.40%,the total methylation rate increased by 16.73%,and the semi-methylation rate increased by 5.57%.When inoculated with the bacteria solution on 12 d,A total of 2298 coexpressed genes,1434 coexpressed up-regulated genes and 769 coexpressed down-regulated genes were detected.The number of upregulated genes was significantly higher than that of down-regulated genes according to the volcanogram.
