摘 要:为探讨种鸡日粮中添加不同水平的精氨酸对海东鸡雏鸡小肠黏膜中营养转运载体(PepT1、SGLT1、GLUT2)及消化酶(APN、SI)基因表达影响,将440只种鸡分为5组,分别添加0.96%、1.16%、1.36%、1.56%和1.76%的L-精氨酸。种鸡饲养44 d,对7日龄雏鸡的十二指肠、空肠、回肠取样,通过实时荧光定量PCR技术对各肠段内相应目的基因进行表达量测定。结果显示:海东鸡不同肠段PepT1、SGLT1、APN mRNA的相对表达量差异极显著(P<0.01),GLUT2和SI mRNA相对表达量存在显著差异(P<0.05);不同L-精氨酸水平海东鸡肠道PepT1、GLUT2、SGLT1、SI mRNA相对表达量存在极显著差异(P<0.01),APN mRNA差异不显著(P>0.05)。由此可见,海东鸡肠道营养转运载体PepT1、SGLT1、GLUT2及消化酶APN、SI mRNA表达具有肠段及L-精氨酸水平的差异性,日粮添加1.16%L-精氨酸对于肠道整体营养转运载体及消化酶的基因表达影响最佳。To investigate the effects of dietary arginine supplementation at different levels on gene expression of nutrient transporters(PepT1, SGLT1, GLUT2) and digestive enzymes(APN, SI) in small intestinal mucosa of Haidong chicken.The 440 breeding chickens were divided into 5 groups, and 0.96%, 1.16%, 1.36%, 1.56% and 1.76%, L-arginine was added to the diet respectively. The chickens were raised for 44 days. The duodenum, jejunum and ileum of 7-day-old chicks were sampled and the corresponding gene expressions in each intestinal segment were measured by real-time fluorescence quantitative PCR. The results showed that the relative expression levels of PepT1, SGLT1 and APN m RNA of duodenum, jejunum, ileum of Haidong chicken were all significantly different(P<0.001), and the relative expression levels of GLUT2 and SI mRNA were significantly different(P<0.05). The relative expressions of PepT1, GLUT2, SGLT1 and SI m RNA at different levels of L-arginine addition were significantly different in Haidong chickens(P<0.001),while there is no difference of APN mRNA(P>0.05). It can be seen that the mRNA expressions of PepT1, SGLT1,GLUT2, digestive enzyme APN and SI of haidong chicken have differences in intestinal segment and L-arginine addition in diet of breeding chickens. Dietary 1.16% L-arginine supplementation in diet of breeding chickens had the best effect on the gene expression of intestinal overall nutrient transporter and digestive enzyme.
