高效液相色谱法同时测定复方磺胺间甲氧嘧啶预混剂中磺胺间甲氧嘧啶及甲氧苄啶的含量

摘　　要：建立了同时测定兽药复方磺胺间甲氧嘧啶预混剂中磺胺间甲氧嘧啶及甲氧苄啶含量的高效液相色谱法。以C18柱为固定相,0.1%磷酸溶液-乙腈(86:14)为流动相,流速1.0 mL/min,柱温40℃,检测波长为240 nm,采用外标法进行定量。磺胺间甲氧嘧啶的线性范围为5.02~100.36μg/mL,回归方程为Area=0.62×Amt-0.428(n=5),r^2=0.9996;甲氧苄啶的线性范围为1.01~20.28μg/mL,回归方程为Area=1.058×Amt-0.075,r^2=0.9995。取复方磺胺间甲氧嘧啶预混剂,按磺胺间甲氧嘧啶和甲氧苄啶含量的100%作添加回收实验,分别作8个平行,测得磺胺间甲氧嘧啶的回收率为98.83%~101.95%,RSD为0.91%;甲氧苄啶的回收率为99.02%~103.09%,RSD为1.23%。该方法简单,准确,精密度好,回收率高,可以用于测量复方磺胺间甲氧嘧啶预混剂的含量。An HPLC method has been developed to determine the content of sulfamonomethoxine and trimethoprim simultaneously in compound sulfamethoxy-diazine premix,using C18 column as stationary phase,with a mobile phase of 0.1%phosphoric solution-acetonitrile(86:14)and wavelength of 240 nm.It was quantified by external standard method and the linear range of sulfamonomethoxine was between 5.02 and 100.36μg/mL.And the regression equation was Area=0.62×Amt-0.428 with the correlation to be more than 0.9996;the linear range of trimethoprim was between 1.01 and 20.28μg/mL,the regression equation was Area=1.058×Amt-0.075 with the correlation to be more than 0.9995.The average recoveries of sulfamonomethoxine from eight spiked premix samples ranged between 98.83%and 101.95%,while relative recoveries of trimethoprim from that ranged between 99.02%and 103.09%.This method was simple and accurate,and had good precision and high recoveries as well,could be used for determination of sulfamonomethoxine and trimethoprim in compound sulfamonomethoxine premix.
