摘 要:塔里木兔是我国新疆地区特有物种,已被列为国家Ⅱ级重点保护动物。本研究基于所测定的塔里木兔线粒体全基因组序列(全长17011 bp),进行了全序列的结构分析,结果表明塔里木兔线粒体基因组包含13个蛋白质编码基因、22个tRNA基因、2个rRNA基因和1个控制区。除了nad6在L链上以外,其他的蛋白质编码基因都在H链上。基于线粒体基因序列,通过生物信息学预测发现除了trnM和trnS1以外,其余tRNA都有经典的三叶草二级结构。线粒体全基因组具有AT偏好性,其结构与目前兔属其他物种线粒体基因组结构类似。并基于塔里木兔和GeneBank已有的12种兔属物种线粒体全基因组序列,分别用ML法和NJ法构建系统进化树,对其系统发生进行了探讨,结果表明塔里木兔与蒙古兔的亲缘关系较近。Yarkand hare(Lepus yarkandensis)is an endemic species in the Xinjiang Uygur Autonomous Region and is listed as a ClassⅡnational protected species.Based on the complete mitochondrial genome sequence(17,011 bp)of Yarkand hare,we analyzed the structure of the mitochondrial genome.The results showed that the mitochondrial genome of Lepus yarkandensis contained 13 protein coding genes,22 tRNA genes,2 rRNA genes and 1 control region.Except for nad6 on the light chain,other protein coding genes were all on the heavy chain.Bioinformatics prediction showed that except trnM and trnS1,the other tRNAs had the classical cloverleaf secondary structure.Mitochondrial genome had AT preference,and its structure was similar to that of other species of Lepus.Based on the complete mitochondrial genome sequences of Lepus yarkandensis and the other 12 species of Lepus,we constructed a phylogenetic tree by use of Maximum Likelihood and Neighbor-Joining methods.The results showed that the relationship between Lepus yarkandensis and Lepus tolai was close.
