摘 要:目的通过对脂肪组织的4种切片处理流程进行比较,筛选出一种比较切实可行的切片处理方法。方法收集2019年中国人民解放军海军第971医院病理科诊断明确的脂肪瘤组织、乳腺脂肪组织和胃、肠、子宫或肾脏器官根治切除术标本中的脂肪组织共298例,分为传统处理法组(A组)、延长浸蜡法组(B组)、丙酮脱脂法组(C组)和改良组织脱水处理流程法组(D组)4组。4组标本分别按照4种不同的方法进行脱水、透明、浸蜡处理,切片最后进行常规苏木精-伊红染色,于光学显微镜下观察,比较各组切片优良率。结果4组切片优良率比较差异具有显著性(χ^2=462.827,P<0.001);C组与D组切片优良率显著高于A组与B组(χ^2=223.666~238.973,P<0.001);而A组与B组之间、C组与D组之间切片优良率比较差异无显著性(P>0.05)。结论采用丙酮脱脂法与改良脱水处理流程法制作的切片优良率高,有利于病理诊断;特别是后者无需额外试剂准备,更省时省力,值得在各医院病理科应用、推广。Objective\To screen out a feasible preparation method for adipose tissue section by comparing the treatment processes for four types of adipose tissue section.\Methods\A total of 298 adipose tissue specimens were collected from lipoma tissue,breast adipose tissue,and the surgical specimens of radical gastrectomy,enterectomy,hysterectomy or nephrectomy after being diagnosed by Department of Pathology in Navy 971 Hospital of The Chinese People’s Liberation Army in 2019.These specimens were divided into conventional treatment group(group A),prolonged wax-embedding group(group B),acetone degreasing group(group C),and modified tissue dehydration group(group D).The four groups of specimens were dehydrated,transpa-rentized,and embedded in wax with the four different methods,respectively,and then HE staining was performed for observation under an optical microscope.The excellent and good rate was compared between groups.\Results\There was a significant diffe-rence in the excellent and good rate of sections between the four groups(χ^2=462.827,P<0.001),groups C and D had a significantly higher excellent and good rate than groups A and B(χ^2=223.666-238.973,P<0.001),while there was no significant difference between group A and group B and between group C and group D(P>0.05).\Conclusion\The sections prepared by acetone degreasing and modified tissue dehydration have a high excellent and good rate and can be used for pathological diagnosis,while for the sections prepared by modified tissue dehydration,the process is time-saving and easy to perform with no additional reagents required and thus holds promise for application in the department of pathology in hospitals.
