摘  要:从暗褐网柄牛肝菌基因组数据中开发高多态性的SSR标记,按照本实验中设计的方法得到43对引物,扩增分型后,30个位点在供试的157个样品中均表现出较高的多态性,其中:三碱基单元的位点22个,四碱基单元的位点4个,五碱基单元的位点1个,六碱基单元的位点3个。PCR产物的长度为114~561 bp。从聚类结果来看,157个样品共有147种多位点分子表型。共享多位点分子表型的样品分别来自同一采样地点,推测来源于同一菌丝无性繁殖系。样品并未按地理群体聚在一起,说明各个地理群体间有基因交流。来自澳大利亚和新西兰的样品也未与来自中国的样品完全分开。SSR markers with high polymorphism were exploited from the genomic data of Phlebopus portentosus.43 primers were obtained in the designed experiment.After PCR amplification and genotyping,30 loci showed higher polymorphism in 157 specimens,of which loci with motifs oftri-,tetra-,penta-and hexa-nucleotides was 22,4,1 and 3 respectively.The length of PCR primers ranged in 114 to 561 bp and the clustering analysis indicated that there were 147 multiple loci molecular phenotypes found in 157 specimens.Specimens shared same genotype from same sampling location,indicating they could be from same mycelia clone underground.There were gene flows in geographic population since clades in the clustering tree didn’t cluster base on geographic location.Samples from Australian and New Zealand didn’t separate from native samples either.
