摘  要:采用蓝粒小麦GLM 1701与4个白粒品种(贵农19、贵农麦30号、中燕96-3和绵麦301)分别组配正反交F1、F2和BC 1F1群体,利用660 K基因芯片和标记筛选对蓝粒性状进行基因定位。结果表明,以蓝粒小麦为父本时,F1籽粒均为浅蓝粒;蓝粒小麦为母本时,F1籽粒均为中蓝粒,说明蓝粒基因为显性,且存在剂量效应。F2分离群体中,糊粉层蓝色和白色的籽粒数目相当,符合5∶4;BC 1F1群体中,正反交F1与白粒亲本进行正反回交,蓝粒白粒比为1∶2,正反交F1与蓝粒亲本进行正反回交,则回交群体全为蓝粒。综合分析回交群体和对F2群体的验证,发现含蓝粒基因的雌或雄配子的传递率均只有67%。通过遗传分析得出,GLM 1701蓝粒性状为一对主效显性基因控制。660 K小麦基因芯片及标记筛选结果显示,控制蓝粒性状的基因位于4 D染色体上,特异性标记Xgwm 165-4 D与目的基因的遗传距离为5.8 cM,本研究将控制蓝粒性状的基因命名为LM 1,且通过T载体克隆得到该标记在中国春的物理位置为412716481~412716679 Mb。In this study the blue-grain wheat GLM 1701 and four white-grain wheat varieties(Guinong 19,Guinongmai 30,Zhongyan 96-3 and Mianmai 301)were used to hybridize by reciprocal cross and acquired F1,F2 and BC 1F1 populations respectively,and the 660-K gene chip and marker screening were used to locate the genes of the blue-grain traits.The results showed that F1 grain was blue when blue-grain wheat was used as male parent,while the F1 grain was medium blue when the blue-grain wheat was the female parent,indicating that blue-grain gene was dominant and had dose effect.In F2 separation population,the number of blue-and white-grains in aleurone layer was the same,corresponding to 5∶4.In BC 1F1 population,the backcross population between the reciprocal cross F1 and white-grain parents showed the ratio of blue grains to white grains being 1∶2,while the backcross population from hybridization of the reciprocal cross F1 with blue-grain parents all appeared blue grains.A comprehensive analysis of the backcross population and validation of F2 population showed that the transmission rate of both male and female gametes containing the blue gene was only 67%.Through genetic analysis,it was concluded that GLM 1701 was controlled by a pair of dominant genes.Meanwhile,the results of 660 K wheat gene chip and marker screening showed that the gene controlling the blue trait was located on the 4 D chromosome,and the genetic distance between the specific marker XGWM 165-4 D and the target gene was 5.8 cM.In this study,the gene controlling the blue-grain trait was named LM1,and the physical location of the marker in Chunchun was 412716481-412716679 Mb after cloning by T vector.
