摘 要:烟酰胺腺嘌呤二核苷酸(NAD^+)是细胞氧化还原反应中的重要辅酶,广泛参与调节细胞生长、分化、能量代谢、凋亡等各种生物学过程。越来越多的研究表明,NAD^+水平与衰老及其相关疾病密切相关。本文综述了以NAD^+为代表的衰老生物标志物,并简述NAD^+的起源、功能与合成,阐述其在衰老进程中的变化和机制,及在衰老表型和衰老相关疾病如寿命、心脏衰老、神经系统退行性疾病、肿瘤和糖尿病中的调控机制和潜在的药物靶点特性,以期为以NAD^+为核心的抗衰老对策研发提供参考。Nicotinamide adenine dinucleotide(NAD^+)is an important coenzyme in cell redox reaction,which is widely involved in the regulation of various biological processes such as cell growth,differentiation,energy metabolism and apoptosis.An increasing number of studies have shown that NAD^+levels are closely related to the progression of aging and age-related diseases.This article reviews the biomarkers of aging represented by NAD^+,and outlines the origin,function and synthesis of NAD^+,and elaborates on the changes and mechanisms of NAD^+in the process of aging.Moreover,this paper expounds the regulatory mechanism and potential drug target characteristics of NAD^+in aging phenotype and age-related diseases such as lifespan,heart aging,neurodegenerative diseases,tumors and diabetes,thus providing reference for the research and development of anti-aging strategies with NAD^+as the core.
