摘 要:该研究采用RACE技术克隆到了膜荚黄芪毛蕊异黄酮葡萄糖苷生物合成途径基因AmIOMT,并进行了生物信息学分析;调查了AmIOMT在膜荚黄芪不同发育时期的根、茎、叶和花中的表达量与芒柄花素之间的关系。结果表明:AmIOMT序列全长为1213 bp,其中,开放阅读框(ORF)编码362个氨基酸多肽,蛋白质分子量约为40.64 kDa,理论等电点pI为4.95,为亲水性蛋白,定位于细胞质中。AmIOMT在根、茎和叶中的表达呈上升趋势,与芒柄花素的含量一致,推测其参与了芒柄花素的生物合成,这为利用生物技术增加芒柄花素的含量提供了理论依据。The Isoflavone O-methyltransferase(IOMT)gene was analyzed Astragalus membranaceus using RACE technique and bioinformatics of AmIOMT was analyzed.The correlations between the expression levels of AmIOMT and the contents of formononetin in root,stem,and leaf were also investigated.The results showed that the full length of AmIOMT was 1213 bp,the open reading frame(ORF)encoded 362 amino acids,the molecular weight of the protein was 40.64 kDa,and the theoretical PI was 4.95,belonged to hydrophilic protein locating in the cytoplasm,respectively.The expression of AmIOMT in roots,stems and leaves showed an upward trend,which was consistent with the content of formononetin.It was speculated that AmIOMT was involved in the biosynthesis of formononetin,which provided a theoretical basis for increasing the content of formononetin by biotechnology.
