摘 要:为建立快速、准确的猪附红细胞体诊断方法,根据GenBank上发布的猪附红细胞体的eno基因序列,设计合成1对特异性引物,通过PCR退火温度筛选出最适温度,将PCR扩增产物进行克隆与测序,并进行特异性试验、敏感性试验及临床应用试验。结果表明:建立的PCR方法扩增出猪附红细胞体eno基因序列大小为170 bp,与GenBank中(序列号AB265823.1)的序列同源性为98%;建立的PCR方法特异性好、敏感性强,具有很好的临床应用价值,该方法为猪附红细胞体病的病原核酸诊断提供了有力支撑。In order to establish a rapid and accurate diagnostic method for Mycoplasma suis,a pair of specific primers were designed and synthesized according to the eno gene sequence of Mycoplasma suis published in GenBank,the PCR products were cloned and sequenced,and the specificity test,sensitivity test and clinical application test were conducted.The results showed that the sequence of eno gene of Mycoplasma suis was 170 bp and the sequence homology with that in GenBank(AB265823.1)was 98%,this method provides a strong support for the diagnosis of swine Mycoplasma suis with pathogenic nucleic acid.
