摘 要:目的观察沙枣叶乙醇提取物对哮喘模型小鼠氧自由基代谢。分析气道炎症及血清白介素4(IL-4)、IgE-免疫球蛋白(IgE)、γ干扰素(IFN-γ)含量的影响,探讨其抑制哮喘气道炎症的作用机制。方法采用改良的卵白蛋白多点注射致敏与雾化吸入激发法建立小鼠哮喘模型,通过测定小鼠肺组织匀浆中超氧化物歧化酶(SOD)、丙二醛(MDA)、一氧化氮(NO)及小鼠血清中IL-4、IgE、IFN-γ含量,镜下观察肺组织病理形态变化及支气管肺泡灌洗液(BALF)中白细胞计数和分类计数。结果与模型组比较,沙枣叶乙醇提取物高、中剂量组SOD含量升高(P<0.05),高、中、低剂量组MDA和NO含量降低(P<0.05);高、中剂量组小鼠血清中IL-4及IgE含量降低(P<0.05),高剂量组IFN-γ含量升高(P<0.05),同时IL-4/IFN-γ值降低(P<0.05);高、中剂量组BALF中白细胞总数以及嗜酸性粒细胞和单核细胞比率降低(P<0.05),中性粒细胞比率升高(P<0.05)。病理切片染色结果显示高、中剂量实验组能够明显减轻肺组织细胞炎性浸润等病理症状。结论沙枣叶乙醇提取物可通过调整体内的氧化与抗氧化系统,降低IL-4及IgE的含量、升高IFN-γ的含量,调节Th1/Th2免疫平衡而改善哮喘模型小鼠气道炎症。Objective To observe the effects of ethanol extract from elaeagnus angustifolia leaves on oxygen free radical metabolism,airway Inflammation,serum interleukin4(IL-4),IgE-immunoglobulin(IgE),interferon-γ(IFN-γ)levels in asthmatic model mice and to explore the mechanism of inhibiting asthma airway inflammation.Methods A mouse asthma model was established by improved ovalbumin multi-point injection sensitization and aerosol inhalation challenge.The contents of superoxide dismutase(SOD),malondialdehyde(MDA),nitric oxide(NO)in lung tissue homogenate and IL-4,IgE,IFN-γin serum of mice were measured.The pathological changes of lung tissue,the white blood cell counts and differential count in bronchoalveolar lavage fluid(BALF)were observed under microscope.Results Compared with the model group,the contents of SOD were increased in the alcohol extract of elaeagnus angustifolia leaves in high and middle dose groups(P<0.05).The contents of MDA and NO were decreased in high,middle and low dose groups(P<0.05).The contents of IL-4 and IgE in serum were decreased in high and middle dose groups(P<0.05),and the contents of IFN-γin serum was increased in high dose groups(P<0.05),and IL-4/IFN-γvalue were decreased(P<0.05).The white blood cell counts and the ratio of eosinophils and monocyte in BALF were increased in high,middle dose groups(P<0.05).Pathological examination showed that the high and middle dose groups could significantly reduce the inflammatory infiltration of lung tissue.Conclusion The alcohol extract of elaeagnus angustifolia leaves could improve the airway inflammation in the asthmatic model mice by adjusting the oxidation and antioxidant system,and it can reduce the content of IL-4 and IgE,and it can increase the content of IFN-γand regulate the immune balance of Th1/Th2.
