摘 要:目的:探讨松杖方对RA患者滑膜细胞(RAFS)凋亡及STAT 3相关蛋白ROR-γt、SOCS-3、pSTAT 3表达的影响。方法:取对数生长期RAFS,分为空白对照组、雷公藤多苷组、来氟米特组、松杖方高、中、低剂量组共六组,分别给以相应含药血清或空白血清干预24 h后,观察细胞形态,流式细胞术分析RAFS的凋亡情况,免疫印迹法检测ROR-γt、SOCS-3、pSTAT 3的表达情况。结果:空白组滑膜细胞持续增殖,各给药组细胞不同程度变形及凋亡。与雷公藤多苷组、来氟米特组比较,松杖方中剂量组RAFS凋亡率相似,松杖方高剂量组凋亡率最高,差异有统计学意义(P<0.05)。与空白对照组比较,各含药血清干预组RAFS中ROR-γt、pSTAT 3的表达量均下降(P<0.05),松杖方高剂量组ROR-γt、pSTAT 3的表达量最低,与雷公藤多苷组及来氟米特组比较差异有统计学意义(P<0.05)。各组RAFS中SOCS-3的表达量差异未见统计学意义(P>0.05)。结论:松杖方可抑制ROR-γt、pSTAT 3的基因表达,抑制炎症信号的传导,促进RAFS凋亡,阻断RA滑膜炎症持续发生。Objective:To investigate the effects of Usnea&Polygonum cuspidatum Formula(UPF)on the apoptosis of synovial cells(RAFS)and the expression of STAT 3 related proteins ROR-γt,SOCS-3 and pSTAT 3 in RA patients.Methods:RAFS was divided into 6 groups:blank control,tripterygium glycosides,leflunomide,and UPF high,medium and low dose,These cells were in the logarithmic growth phase.After 24 h intervention with corresponding medicated serum or blank serum,the cell morphology was observed by an inverted microscope,apoptosis was analyzed by flow cytometry,and the expression of ROR-γt,SOCS-3,and pSTAT 3 was detected by western blotting.Results:Synovial cells in the blank group continued to proliferate,and the morphological deformation and apoptosis of the cells in each treatment group increased to different degrees.The apoptotic rate of the middle-dose group of UPF was similar to that of the tripterygium glycoside group and the leflunomide group.The high-dose group of UPF was higher than these three groups and was statistically significant(P<0.05).The expression levels of ROR-γt and pSTAT 3 in RAFS in each intervention group were lower than those in the blank control group(P<0.05).The expression of ROR-γt and pSTAT 3 in the high-dose UPF group was the lowest,which was significantly different from the tripterygium glycosides and leflunomide group(P<0.05).There was no significant difference in the expression of SOCS-3 in each group(P>0.05).Conclusion:UPF be related to decreased expression of ROR-γt and pSTAT 3 protein,poor inflammatory signal conduction,and increased RAFS apoptosis may,which may cause the continuous occurrence of RA synovial inflammation and be cut off.
