印度橄蝗线粒体基因组测定及系统发育分析

摘　　要：从分子水平分析印度橄蝗(Tagasta indica)与近缘昆虫的系统发生关系,采用一代PCR测序方法对印度橄蝗的线粒体基因组进行测序,对序列进行拼接、注释和特征分析,并利用线粒体基因组的13个蛋白编码基因(PCGs)和2个核糖体RNA(rRNAs)构建贝叶斯(BI)树。结果表明,印度橄蝗的线粒体基因组长度为15531 bp,包括13个PCGs、22个转运RNA(tRNAs)、2个rRNAs和1个控制区;线粒体基因排列紧凑,基因排序与祖先序列相同。线粒体蛋白编码基因中,除nad2、cox1和nad6为非标准起始密码子GTG、ACT和TTG外,其余均为标准起始密码子ATN;终止密码子中,除cox3为不完全终止密码子TA外,其余均为完整终止密码子TAA或TAG。线粒体的22个tRNAs中,除trnSAGN缺少DHU臂外,其余均为典型三叶草结构。错配方式主要为G-U错配。构建的贝叶斯树支持瘤锥蝗科(Chrotogonidae)的单系性。To analyze phylogenetic relationship between Tagasta indica and related insects at molecular level,the first sequencing method of PCR was used to determine mitochondrial genome of T.indica.Sequences were jointed,noted and analyzed,and Bayesian tree was constructed with 13 protein coding genes(PCGs)and 2 ribosomal RNAs(rRNAs)of mitochondrial genome.Results showed that mitochondrial genome of T.indica was 15531 bp in length,including 13 PCGs,22 transport RNAs(tRNAs),2 rRNAs and one control region.Mitochondrial genes were compactly arranged,and the gene order was identical to the ancestral sequence.Most of mitochondrial protein coding genes started with typical ATN codons,except for nad2,cox1 and nad6,which begined with GTG,ACT and TTG,respectively.Stop codons were complete termination TAA or TAG,except for cox3,which stopped with TA.22 tRNAs were well folded into a clover-leaf secondary structure,except for trnSAGN,which lacked DHU stem.The mismatch was mainly G-U mismatch.Bayesian inference supported the monophyla of Chrotogonidae.
