摘 要:目的研究孕哺期染铅对仔鼠海马突触发育的损伤以及硒的拮抗作用。方法从母鼠受孕第1d起给予醋酸铅水溶液[1g/(kg·bw),每2d一次]灌胃,硒干预组给予含亚硒酸钠的饮水(400μg/L和1600μg/L)自由饮用。仔鼠出生后21d后取脑,用高尔基染色法观察海马突触的发育,进行树突棘计数;用RT-PCR检测突触可塑性相关基因Drebrin、PSD95和synaptophysin的mRNA表达,分析硒对铅毒性的拮抗作用。结果孕哺期母鼠染铅导致仔鼠体质量下降、海马神经突触树突棘密度降低,突触可塑性相关基因Drebrin和PSD95的mRNA表达显著下降;硒的干预能够明显增加仔鼠体质量和海马树突棘密度,但Drebrin和PSD95的mRNA表达未见明显增加。结论在一定剂量下给予孕哺期母鼠补硒能够拮抗铅对仔鼠的毒性作用,具体机制有待进一步研究。Objective To study the effects of lead staining on hippocampal synapse development and selenium antagonism in young rats during gestational feeding period.Methods Lead acetate solution[1g/(kg·bw),once every two days]was fed to pregnant mice by intragastric administration since the first day of conception.Drinking water containing sodium selenite(400μg/L and 1600μg/L)was given to the selenium intervention group freely.Brains of 21-days old progeny mice were used for further study.Development of hippocampal synapses was observed by Golgi staining,and the dendritic spine was counted.The expression of mRNA was detected by RT—PCR for the hippocampal synaptic plasticity-related genes including Drebrin,PSD95 and synaptophysin.Antagonistic effect of selenium to lead toxicity was analyzed.Results Lead exposure during pregnancy and lactation causes damages to progeny mice such as decrease of body weight,reduction of dendritic spine density and mRNA expression of Drebrin and PSD95.Selenium intervention significantly increases the body weight and dendritic spine density,but does not affect the mRNA expressions of Drebrin,PSD95 and synaptophysin.Conclusion Certain dose of selenium could antagonize the toxicities induced by lead during pregnancy and lactation,still the mechanism is unclear and needs further elucidation.
