摘 要:为了确定内蒙古额济纳旗边境地区蜱虫种类以及其保守基因16SrRNA是否具有基因多态性。本试验选取该地区3个代表性采样点,收集双峰驼的体表寄生蜱虫383只。通过传统形态学方法,保守基因16S rRNA和线粒体细胞色素C氧化酶亚基Ⅰ(COⅠ)基因PCR序列扩增、测序、系统发育进化树分析进行蜱虫种类鉴定。通过Genetyx-Version 6软件对每个采样点随机选取的10个样本进行16S rRNA序列基因多态性分析。结果显示:本试验所采集的蜱虫形态均符合亚洲璃眼蜱的典型形态特征;经16S rRNA基因和COⅠ基因的序列绘制出的系统发育进化树结果显示该蜱与我国新疆地区的亚洲璃眼蜱在同一个进化分支上,并能够很好的聚类;在蜱虫的16S rRNA基因多态性的结果中发现,只有3个序列与其他的27个序列表现出不同程度的基因多态性,说明亚洲璃眼蜱中存在基因多态性的变化。这为以后蜱虫的鉴定以及探究基因多态性方面提供参考。To determine the species of ticks and the polymorphism of 16S rRNA gene of them in Ejina Banner of Inner Mongilia,the border area of China,a total of 383 ticks were collected from Bactrian camels at three sampling regions selected randomly in this area.Then,they were identified with traditional morphological observation method and screened by phylogenetic analysis of 16S rRNA and mitochondrial cytochrome Coxidase subunit Ⅰ(CO Ⅰ)gene,respectively.In addition,the sequence polymorphism of the 16S rRNA gene in 10 randomly selected samples from each sampling regions was analyzed by using Genetyx-Version 6 software.The results indicated that all the collected ticks had the typical morphological characteristics of Hyalomma asiaticum,and the phylogenetic analyses based on 16S rRNA and CO Ⅰ gene demonstrated that these ticks were clustered perfectly with H.asiaticum isolated in Xinjiang of China.Moreover,the results of gene polymorphism of 16S rRNA showed that only three isolates showed different degrees of gene polymorphism compared with the other 27 isolates,indicating that there was genetic polymorphisms in H.asiaticum.This study provides a reference for the identification of ticks and the exploration of gene polymorphism in the future.
