哺乳仔猪自然感染猪流行性腹泻病毒病例的病理形态学观察

摘　　要：为观察自然状态下猪流行性腹泻病毒(porcine epidemic diarrhea virus,PEDV)感染哺乳仔猪的病理变化,以某猪场7头确定为PEDV感染的哺乳仔猪为研究对象,观察其临床症状、剖检变化、组织病理学变化、亚细胞水平结构变化及病毒的组织分布情况。结果显示,感染仔猪消瘦、呕吐、水样腹泻。剖检发现,小肠鼓气,肠壁变薄;胃内有未消化的乳凝块;肠系膜淋巴结充血肿胀。组织病理学结果显示,肠组织普遍出现绒毛萎缩变短;肠系膜淋巴结和脾中淋巴细胞减少;心肌局灶性坏死等。亚细胞结构观察发现,空肠黏膜上皮微绒毛稀疏,上皮细胞胞质空泡化,内质网缢断,线粒体肿胀变圆,并可见散在的冠状病毒粒子。免疫组化结果显示,除直肠外,肠道黏膜上皮细胞胞质中均发现阳性信号,偶尔在肠腺细胞胞质内亦可见到。结果表明,在PEDV自然感染条件下,哺乳仔猪组织器官出现了典型的病理变化,与人工感染相比,致病机理与发病规律大致相同,也存在一定的差别。To observe the pathological changes of suckling piglets naturally infected with porcine epidemic diarrhea virus(PEDV),seven piglets infected with PEDV in a pig farms were selected as the research object.The clinical symptoms,autopsy changes,histopathological changes,structural changes at the subcellular level,and tissue distribution of virus were observed.The results showed that the infected piglets exhibited symptoms of weight loss,vomiting,and watery diarrhea.Dissection examination revealed that the small intestine was inflated,and the intestinal wall became thin,there were undigested milk clots in the stomach,and mesenteric lymph nodes were congested and swollen.Histopathological results showed that atrophy and shortening of villus are common in intestinal tissues,and the number of lymphocytes in spleen and mesenteric lymph nodes decreased,and focal myocardial necrosis occurred.The results of subcellular structure observation found that the microvilli on jejunal epithelial cells was sparse,and the cytoplasm was vacuolated,the endoplasmic reticulum was broken,the mitochondria were swollen and round,and the scattered coronavirus particles are visible in the cytoplasm.Immunohistochemical results showed that,in addition to the rectum,positive signals were found in the cytoplasm of intestinal mucosal epithelial cells,and occasionally in the cytoplasm of intestinal gland cells.The results showed that,in the case of natural infection of PEDV,the tissues and organs of suckling piglets showed typical pathological changes.The pathogenic mechanism and pathogenesis of natural infection are similar to that of artificial infection in suckling piglets,but there are some differences between them.
