摘 要:甘蔗是雌雄同株高度杂合的异源多倍体植物。为去除杂交育种过程中因母本去雄不彻底导致的自交现象的干扰,有必要对杂交后代进行杂种真实性鉴定。本研究以‘新台糖25号’为母本、‘云蔗89-7’为父本进行有性杂交构建大规模遗传群体,获得F1代实生苗共17923株。从54个甘蔗SSR标记中筛选出一个具有父本特异性的SSR标记SMC31CUQ,通过多态性序列分析,重新设计了可特异性检测父本序列的PCR引物对SMC31CUQ-F/SMC31CUQ-1-R,并用其对整个遗传群体实生苗进行PCR鉴定,发现其中有9310株具有该父本特异性SSR标记,占群体的51.94%。大规模真杂种的获得,为进一歩构建甘蔗特定农艺性状亚群体奠定了基础。Sugarcane is a monoecious and highly heterozygous allopolyploid crop.In order to eliminate the interference of selfing caused by incomplete emasculation,It is necessary to identify the authenticity of hybrid progeny.In this study,"ROC"25 was used as a female parent and Yunzhe89-7 was used as a male parent,their F1 generation was obtained by hybridization.A large-scale F1 generation population containing 17923 seedlings was obtained.A total of 54 sugarcane SSR markers had been screened,and one of the markers,SMC31CUQ,was identified to be paternal specific.A specific PCR primer pair SMC31CUQ-F/SMC31CUQ-1-R was designed from the unique amplicon among the polymorphism fragments generated by SSR primer pair SMC31CUQ through discrimination of the shared sequences.This newly designed primer pair allowed the identification of male parent and offspring that carried the male-specific marker,and equivalent of SSR generated by marker SMC31CUQ.PCR identification of the whole genetic population revealed that 9310 progenies(51.94%)carried the male paternal specific SSR marker.These largescale true hybrids greatly facilitate the effort to construct sugarcane subpopulation with varied agronomic traits.
