摘 要:该实验旨在通过对发酵条件进行优化,提高链霉菌PTY-3绿色素的产量。以分离自吉林省四平市大棚黑土中的一株链霉菌PTY-3为实验菌株,对发酵时间、装液量、接种量、发酵起始pH和发酵温度进行了优化。结果表明,在发酵时间8 d,发酵温度为28℃,接种量为10%,装液量110 m L/250 mL和发酵起始pH值为7.5的条件下培养时绿色素的色价最高,可达77.0 U/m L,是未优化前(57.3 U/mL)的1.34倍。该研究为天然绿色素的进一步工业化生产提供了科学依据。This experiment aims to increase the yield of green pigment by optimizing the culture condition of the green pigment produced by Streptomyces sp.PTY-3,which was isolated from the black soil of Siping City,Jilin Province.Based on the single factor experiment,fermentation time,bottle volume,inoculation volume,initial pH and fermentation temperature as five single factors were selected and optimized by orthogonal test.The results showed that the optimum conditions of fermentation was as follows:fermentation time 8 days,fermentation temperature 28℃,liquid volume 110mL,inoculum size 10%,the initial pH 7.5.Under these conditions,the highest green pigment reached 77.0 U/mL,corresponding to1.34 times over the standard conditions before optimization(57.3 U/mL).This study provides a theoretical basis for the industrial production of green pigment.
