摘　　要：目的:探讨obestatin对大鼠胃和十二指肠运动的影响。方法:选取清洁级健康成年Wistar大鼠30只,体质量200~250克,采用随机数字表法分为对照组和obestatin 15.00×10^-8mol/L组,每组15只,禁食18 h以上,观察静脉注射obestatin对禁食大鼠胃肠运动的影响。另外选取32只Wistar大鼠,随机分为4组,分别为对照组,obestatin 3.75×10^-8mol/L组、7.50×10^-8mol/L组、15.00×10^-8mol/L组,禁食18 h以后进食3 g食物,观察不同浓度obestatin对进食状态下大鼠胃肠运动的影响。用RM-46胃运动记录仪连续描记胃运动曲线。结果:静脉注射obestatin后对禁食状态下胃窦或十二指肠中III期样收缩的频率与注射生理盐水的对照组相比差异无统计学意义(P>0.05)。禁食18 h以后进食3 g食物,静脉注射obestatin时,十二指肠开始III期样收缩的时间延长。十二指肠开始III期样收缩的时间依赖注射obestatin的浓度,obestatin的浓度越大,十二指肠开始III期样收缩的时间越晚,差异具有统计学意义(P<0.05)。在静脉注射obestatin后30~90 min内,7.50×10^-8mol/L组、15.00×10^-8mol/L组与对照组相比胃运动变化率降低(P<0.05)。结论:Obestatin可抑制进食状态下的胃十二指肠蠕动,但在禁食状态下则不起作用。Objective:To investigate the effects of obestatin on gastric and duodenum motility in rats.Methods:Thirty pathogen-free healthy adult Wistar rats,weighing 200-250 g,were divided into control group and obestatin 15.00×10^-8mol/L groups using a random number table,15 cases in each group.After fasting for more than 18 hours,the effects of obestatin on gastrointestinal motility were observed.32 pathogen-free healthy adult Wistar rats were divided into control group and obestatin 3.75×10^-8mol/L group,7.50×10^-8mol/L group and 15.00×10^-8mol/L group,8 cases in each group.After fasting for 18 hours,3 g food was consumed to observe the effects of obestatin at different concentrations on gastrointestinal motility in rats.The gastric motion curves were recorded continuously by RM-46 gastric motion recorder.Results:There was no significant difference in the frequency of phase III like contractions in the gastric antrum or duodenum after intravenous administration of obestatin as compared with the control group(P>0.05).After fasting for 18 hours,3 g food was taken in rats,and the time of duodenal phase III like contraction was prolonged when obestatin was injected intravenously.The start time of duodenal phase III like contraction was dependent on the concentration of obestatin injected intravenously.The higher the concentration of obestatin,the later the start time of duodenal phase III like contraction,with statistically significant(P<0.05).The change rate of gastric motility was significantly lower in 7.50×10^-8mol/L group and 15.00×10^-8mol/L group than that in the control group within 30-90 min after intravenous administration of obestatin(P<0.05).Conclusion:Obestatin can inhibit the peristalsis of gastroduodenum in the feeding state,but it has no effect on fasting.
