摘　　要：目的:制备雷公藤甲素固体脂质纳米粒(triptolide-solid lipid nanoparticles,TPL-SLN),并考察其抗慢性皮炎湿疹的作用。方法:以Compritol 888 ATO为油相、Tween 80为表面活性剂、大豆卵磷脂和Transcutol HP为助表面活性剂,通过处方筛选,制备TPL-SLN。以不同浓度的2,4-二硝基氯苯丙酮溶液为致敏剂和激发剂,建立小鼠耳部慢性皮炎湿疹模型,考察TPL-SLN对慢性湿疹造成的肿胀症状以及对血清中细胞因子分泌的影响。结果:本研究制备的TPL-SLN粒径为(90.69±1.82)nm,且分布均匀,能有效抑制小鼠皮炎湿疹造成的耳肿胀症状,可显著降低皮肤炎性因子IFN-g、IL-4的表达。结论:本研究制备的TPL-SLN能够有效改善小鼠皮炎湿疹症状,有望为雷公藤甲素治疗慢性皮炎湿疹提供一种新的给药策略。Objective:To prepare triptolide-solid lipid nanoparticles(TPL-SLN)and investigate their effect on chronic dermatitis and eczema.Methods:Triptolide-solid lipid nanoparticles were prepared using Compritol 888 ATO as the oil phase,Tween 80 as the surfactant,soybean lecithin and Transcutol HP as the co-surfactant.A mice model with chronic ear dermatitis and eczema was established by sensitizing and stimulating using 2,4-dinitrochlorobenzene solution with different concentration to investigate their effect on swelling,skin redness and the serum concentrations of IFN-γ,IL-4 and IL-8.Results:The prepared TPL-SLN had a particle size of 90.69±1.82 nm and a uniform distribution,and could effectively inhibit the ear swelling,erythema and other symptoms caused by dermatitis and eczema in mice and significantly reduce the expression of skin inflammatory factors IFN-and IL-4.Conclusion:The TPL-SLN can effectively improve the symptoms of dermatitis and eczema in mice and is expected to provide a new delivery strategy for triptolide for the treatment of chronic dermatitis and eczema.
