摘 要:为了研究不同激素配比下杨树分化及生根生长情况,筛选最适合山西省杨树的组培体系,试验通过对比不同植物激素配比条件下杨树诱导愈伤组织及生根情况,分析幼苗长势及生理指标变化,研究杨树组培幼苗的炼苗过程。结果表明,培养基体系为MS+0.05 mg/L NAA+0.08 mg/L TDZ时,40 d愈伤组织诱导出芽数最高,单位叶片出芽率为4.4;培养基体系为1/2 MS+0.01 mg/L IBA+0.01 mg/L NAA时,7 d生根数量最多,单株初级根数量为4.2;培养基体系为1/2 MS+0.10 mg/L IBA+0.01 mg/L NAA时,40 d杨树幼苗长势最好。另外,炼苗试验结果显示,组培幼苗水培14 d后移栽至土壤基质中并保湿13 d可以显著提高幼苗成活率。不同激素配比条件下,诱导杨树分化及生根存在差异,且影响后期幼苗长势及生理状况。The purpose of this study was to study the differentiation and rooting of poplar with different hormone ratios,and to screen the optimal tissue culture system of poplar trees in Shanxi province.Callus and rooting induced of poplar under different plant hormone ratios were compared to analyze the growth and physiological index changes of the seedlings and study the process of tissue culture of poplar seedlings.The results showed that when MS+0.05 mg/L NAA+0.08 mg/L TDZ was used in the medium system,the number of induced buds of callus at 40 d was the highest,and the budding rate per unit leaf was 4.4.When the culture medium was 1/2 MS+0.01 mg/L IBA+0.01 mg/L NAA,the number of rooting was the highest at 7 d,and the number of primary roots per plant was 4.2.Poplar seedlings showed the best growth at 40 d when the medium system was 1/2 MS+0.10 mg/L IBA+0.01 mg/L NAA.In addition,the results of the seedling training experiment showed that the survival rate of the tissue cultured seedlings could be significantly improved after 14 d of hydroponics and moisturizing 13 d.In conclusion,different hormone ratio induced different differentiation and rooting of poplar,and affected seedling growth and physiological status.
