摘  要:为了探讨不同浓度的胰岛素对体外培养的鸡胚盲肠上皮细胞生长和增殖的影响,试验选用16日龄SPF鸡胚,采用嗜热菌蛋白酶消化法无菌分离鸡胚盲肠组织,以含不同质量浓度胰岛素(0、5、10、15、20μg/mL)的培养基对盲肠上皮进行体外培养,对细胞克隆形成率、生长曲线和上皮细胞角蛋白18进行测定。结果表明,培养基中添加10μg/mL胰岛素时,鸡胚盲肠上皮细胞的增殖差异极显著,胰岛素质量浓度为15、20μg/mL时,鸡胚盲肠上皮细胞增殖略有升高,但与胰岛素10μg/mL组相比,细胞增殖差异不显著;角蛋白18鉴定结果为阳性。表明在体外鸡胚盲肠上皮细胞培养中,10μg/mL胰岛素能促进鸡胚盲肠上皮细胞分裂、生长和增殖。胰岛素作为一个体外细胞生长的关键因子,可促进鸡胚盲肠上皮细胞的生长和增殖。To investigate the effects of different insulin concentrations on the growth and proliferation of chicken embryo cecum epithelial cells cultured in vitro,16-day-old SPF chicken embryos were selected for aseptic separation of chicken embryo epithelium by protease digestion of thermophilic bacteria in this study.The cecum epithelium was cultured in medium containing different insulin concentrations(0,5,10,15,20μg/mL),and the cell clone formation rate,growth curve and epithelial cytokeratin 18 were measured.The results showed that the proliferation of chicken embryo cecum epithelial cells had significantly different when 10μg/mL insulin was added to the culture medium.At 15,20μg/mL of insulin,the proliferation of chicken embryo cecum epithelial cells increased slightly,but there was no significant difference in cell proliferation compared with that of 10μg/mL insulin group.Cytokeratin 18 was identified as positive.The study showed that 10μg/mL insulin could promote the growth and proliferation of chicken embryo cecum epithelial cells in vitro.As a key factor for cell growth in vitro,insulin promotes the growth and proliferation of chicken embryo cecum epithelial cells.
