摘 要:目的基于网络药理学方法研究野菊花主要抗菌活性成分、靶点,初步探讨其作用机制。方法通过中药系统药理学数据库(TCMSP)、GeneCards数据库筛选出野菊花主要抗菌活性成分、靶点,采用Cytoscape 3.7.2软件构建可视化的“药物-成分-靶点-疾病”网络,依据在线STRING数据库构建蛋白互作网络(PPI),应用R软件对关键靶点进行GO及KEGG通路分析。结果野菊花抗菌的潜在靶点有19个,蛋白互作网络提示IL-1β、CXCL8、IL-10、ICAM1、IFNγ、CASP3等可能是野菊花抗菌的核心靶点。GO分析结果显示,野菊花抗菌潜在基因的生物功能主要涉及细胞因子受体结合、细胞因子活性、受体配体活性、受体调节活性等;KEGG结果显示,野菊花抗菌潜在基因的通路主要涉及甲型流感病毒(Influenza A)、IL-17信号通路(IL-17 signaling pathway)、南美锥虫病(Chagas disease)、Toll样受体信号通路(Toll like receptor signaling pathway)、TNF信号通路(TNF signaling pathway)、弓形虫病(Toxoplasmosis)等信号通路。结论该研究通过网络药理学筛选,解释了野菊花抗菌活性的作用机制,为后续深入开展其抗菌作用研究奠定了基础。Objective To study the antibacterial active compounds and predict targets of Flos Chrysanthemi Indici,and to explore the mechanism through network pharmacology.Methods Antibacterial active compounds and targets of Flos Chrysanthemi Indici were screened on Traditional Chinese Medicine Systems Pharmacology(TCMSP)database and Gene cards databases.Cytoscape 3.7.2 software was used to construct the drug-component-target-disease network diagram.Online STRING database was used to draw protein-protein interaction network(PPI).Gene ontology(GO)enrichment analysis and KEGG pathway enrichment analysis on key targets were performed through R software.Results There were 19 potential targets for the antibacterial activity of Flos Chrysanthemi Indici.The protein interaction network suggested that IL-1β,CXCL8,IL-10,ICAM1,IFNγ,and CASP3 may be the core targets.GO analysis showed that the biological functions of potential genes of antibacterial activity of Flos Chrysanthemi Indici were mainly related to cytokine receptor binding,cytokine activity,receptor-ligand activity,receptor regulator activity,et al.KEGG pathway enrichment results showed that the pathways of potential genes of antibacterial activity of Flos Chrysanthemi Indici mainly involved Influenza A,IL-17 signaling pathway,Chagas disease(American trypanosomiasis),Toll-like receptor signaling pathway,TNF signaling pathway,and Toxoplasmosis signaling pathways.Conclusion The research explained the antibacterial mechanism of Flos Chrysanthemi Indici based on the network pharmacology screening and lay a foundation for further study of its antibacterial effect.
