摘 要:目的:探讨电针通过缓解2型糖尿病大鼠氧化应激状态,保护骨骼肌细胞线粒体正常形态结构,从而改善胰岛素抵抗、降低血糖的机制。方法:选取14只Zucker糖尿病肥胖大鼠(ZDF)及7只非糖尿病大鼠(ZL)为研究对象。将成模后ZDF大鼠随机分为模型组、电针组,每组各7只,ZL大鼠为空白组,干预4周,每7 d测其空腹血糖。4周后取血清,以放射免疫法检测血清活性氧(ROS)、丙二醛(MDA)、总抗氧化能力(T-AOC)及超氧化物歧化酶(SOD)水平,并计算胰岛素抵抗指数(HOMA-IR)。结果:与空白组比较,模型组血糖、ROS、MDA及HOMA-IR显著升高(P <0.05),T-AOC、SOD显著下降(P <0.05);与模型组比较,电针组血糖明显改善(P <0.05),T-AOC升高(P <0.05),MDA、ROS下降(P <0.05);通过均值比较分析,与模型组比较,电针组SOD升高,HOMA-IR降低;通过电镜观察,模型组线粒体结构损伤明显,电针组受损程度小。结论:电针可通过降低ROS和MDA、增加SOD和T-AOC水平减少OS,从而保护骨骼肌线粒结构,进而降低HOMA-IR、改善IR以控制2型糖尿病大鼠血糖。Objective: To explore the mechanism of electro-acupuncture in improving insulin resistance( IR)and lowering blood glucose by relieving oxidative stress and protecting the normal morphological structure of mitochondria in skeletal muscle cells in rats with type 2 diabetes mellitus( T2DM). Methods: 14 Zucker diabetic fatty rats( ZDF) and 7 non-diabetic rats( Zucker lean,ZL) were selected as the study subjects. ZDF rats were randomly divided into the model group and the electro-acupuncture group,with 7 rats in each group;ZL rats were taken as the blank control. The rats were treated for 4 weeks and fasting blood glucose was measured every 7 days. After 4 weeks of intervention,serum levels of ROS,MDA,T-AOC and SOD were measured by radioimmunoassay method. HOMA-IR was calculated as well. Results: Compared to those in the blank group,blood glucose,ROS,MDA,and HOMA-IR were significantly increased and T-AOC and SOD were significantly decreased in the model group( P < 0. 05). Compared to those in the model group,blood glucose was significantly improved,the serum level of T-AOC was increased and the serum levels of MDA and ROS were decreased in the electro-acupuncture group( P < 0. 05). Compared to those in the model group,the serum level of SOD was increased and serum of HOMA-IR was decreased in the electro-acupuncture group( P < 0. 05). Electron microscopy showed that the mitochondrial structure damage of the model group was obvious and it was lightly damaged in the electro-acupuncture group. Conclusion: Electro-acupuncture can reduce OS by decreasing ROS and MDA,increasing SOD and T-AOC,thereby protecting skeletal muscle mitochondrial structure,reducing HOMA-IR and improving IR to control blood glucose in rats with T2DM.
