摘 要:目的:观察不同频率电针内关穴对大鼠心肌缺血再灌注损伤的保护效应及对心肌组织中M2AChR、α7nAChR的影响。方法:将32只大鼠随机分为假手术组、模型组、低频电针组和高频电针组,采用结扎冠状动脉前降支(LAD)的方法制备模型,电针刺激于术前7 d进行。观察心电图(ECG) ST段幅值的变化,并检测大鼠血清中CK-MB、LDH的水平,检测大鼠左室心肌组织中M2毒蕈碱M2受体(M2AChR)、α7烟碱受体(α7nAChR)的表达。结果:低频电针组结扎后30 min的心电图ST段、血清CK-MB和LDH的水平均较模型组显著降低(P <0.01或P <0.05),左室心肌组织中M2AChR、α7nAChR的表达显著增加(P <0.01或P <0.05)。高频电针组未观察到上述效应。结论:低频电针预处理能够减轻心肌缺血再灌注损伤,并可上调心肌组织中M2AChR、α7nAChR的表达。Objective: To observe the effect of electro-needling Neiguan( PC6) with different frequency on myocardial ischemia-reperfusion injury( MIRI) in rats and its influence to the expressions of M2AChR and α7nAChR in the left ventricular tissue. Methods: 32 rats were randomly divided into the sham operation group,the model group,the low-frequency electro-acupunture group and the high-frequency electro-acupucture group. Ligation of LAD was adopted to establish the model,and electro-acupuncture was conducted for 7 days before the operation. The ST amplitudes of ECG,the serum levels of CK-MB and LDH and the expressions of M2AChR and α7nAChR in the left ventricular tissues were observed and detected. Results:Compared to those in the model group ST amplitudes of the ECG were decreased significantly 30 min after ligation( P < 0. 01),and the serum levels of CK-MB and LDH were reduced significantly in the low-frequency electro-acupunture group( P < 0. 05). The expressions of M2AChR and α7nAChR were increased significantly in the left ventricular tissues( P < 0. 05,P < 0. 01) in the low-frequency electro-acupuncture group. But all above effects were not observed in the high-frequency electro-acupuncture group. Conclusion: Low-frequency electro-needling PC6 can alleviate the myocardial ischemia-reperfusion injury and up-regulate the expressions of M2AChR and α7nAChR in the left ventricular tissues.
