摘  要:目的观察潜阳合剂对血管紧张素Ⅱ(AngⅡ)诱导的心肌细胞β-肌球蛋白重链(MYH7)、骨骼肌肌动蛋白α1(ACTA1)、脑钠肽(BNP)mRNA及蛋白表达的影响,探讨潜阳合剂改善心肌纤维化的机制。方法建立AngⅡ诱导的心肌细胞异常增殖的细胞模型,并采用潜阳合剂干预。用CCK-8法测定潜阳合剂对心肌细胞的细胞毒性和拮抗AngⅡ作用的最佳浓度,并分成空白组、AngⅡ组(800 nmol/L)、AngⅡ+潜阳合剂组(稀释4000倍),应用蛋白质免疫印迹法(Western Blot)检测MYH7、ACTA1及BNP蛋白表达,实时荧光定量聚合酶链式反应(q-PCR)检测加入潜阳合剂后MYH7、ACTA1、BNP mRNA表达。结果与空白组比较,AngⅡ组MYH7、ACTA1及BNP mRNA及蛋白表达明显上升(P<0.05或P<0.01);与AngⅡ组比较,AngⅡ+潜阳合剂组MYH7、ACTA1、BNP mRNA及蛋白表达明显下降(P<0.05或P<0.01)。结论潜阳合剂可以通过降低MYH7、ACTA1、BNP mRNA及蛋白的表达起到改善心肌纤维化的作用。Objective To observe the effect of Qianyang Decoction(QYD)onβ-myosin heavy chain gene(MYH7),actinα1(ACTA1),brain natriuretic peptide(BNP)mRNA and protein expression,and explore the mechanism of QYD improving myocardial fibrosis.Methods The cell model of abnormal appreciation of myocardial cells induced by angiotensinⅡ(AngⅡ)was established,which was intervened by QYD.The cytotoxicity of myocardial cells and the optimal concentration of QYD to antagonize the effect of AngⅡwere tested by CCK-8.The cell models of abnormal appreciation of myocardial cells were divided into blank group,AngⅡgroup(800 nmol/L),and AngⅡ+QYD group(diluted 4000 times).The protein display of MYH7,ACTA1,and BNP after adding QYD was analyzed by Western Blot.ThemRNA indexes of MYH7,ACTA1,and BNP after adding QYD were detected byquantitative polymerase chain reaction(q-PCR).Results Compared with blank group,the protein display of MYH7,ACTA1,and BNP in AngⅡgroup significantly increased(P<0.05 or P<0.01);Compared with AngⅡgroup,the expression of MYH7,ACTA1,BNP mRNA,and protein expression in AngⅡ+QYD group significantly decreased(P<0.05 or P<0.01).Conclusion Qianyang Decoction can improve fibrosis of cardiomyocytes by reducing the mRNA and protein expression of MYH7,ACTA1,and BNP.
