摘 要:目的观察加入逆转剂量米非司酮(MIF)的人上皮性卵巢癌耐药细胞系SKOV3/DDP耐药情况。方法取对数生长期的人上皮性卵巢癌细胞系SKOV3、SKOV3/DDP细胞,MTT法观察MIF对SKOV3、SKOV3/DDP细胞增殖的影响,测算细胞增殖抑制率,筛选MIF的非细胞毒性剂量(以此为逆转剂量)。MTT法测算SKOV3/DDP细胞对顺铂(DDP)的耐药指数(RI)。取适量SKOV3/DDP细胞,分为A、B两组,A组加入RI剂量的的DDP,B组加入RI剂量的的DDP+5μmol/L的MIF,分别于培养24、48、72 h时MTT法测算细胞IC50,计算MIF对SKOV3/DDP细胞的耐药逆转倍数(RF)。利用流式细胞术(FCM)检测加入5μmol/L MIF后SKOV3/DDP后的细胞周期。结果不同浓度的MIF均可抑制SKOV3、SKOV3/DDP的增殖,且呈剂量依赖性。MIF对SKOV3/DDP的逆转剂量为5μmol/L。SKOV3、SKOV3/DDP对DDP的IC50分别为3.22、14.83μg/mL,SKOV3/DDP对DDP的RI为4.61。培养24、48、72 h时A组IC50分别为(23.13±0.83)、(14.83±0.52)、(10.07±0.56)μg/mL;B组分别为(17.06±0.53)、(7.77±0.47)、(4.06±0.69)μg/mL,MIF对SKOV3/DDP细胞耐药性的RF分别为1.36、1.91、2.48。培养24、48、72 h时,加入5μmol/L MIF的SKOV3/DDP细胞G0/G1期所占比例逐渐增加(48.7%、53.9%、55.1%、59.0%),S期所占的比例则逐渐减小(13.5%、9.6%、7.7%、5.8%)。结论加入5μmol/L的MIF可抑制SKOV3/DDP细胞增殖,5μmol/L的MIF可减轻SKOV3/DDP细胞对DDP耐药性。MIF减轻SKOV3/DDP细胞对DDP的耐药性机制可能为MIF将SKOV3/DDP细胞周期阻滞于G0/G1期。Objective To observe the drug resistance of human epithelial ovarian cancer cell line SKOV3/DDP added with reversal dose of mifepristone(MIF).Methods The proliferation inhibition rate of MIF on SKOV3 and SKOV3/DDP cell in logarithmic growth phase were detected by MTT method,and we selected the noncytotoxic dose of MIF as reversal dose.The resistance index(RI)of SKOV3/DDP cells to cisplatin(DDP)was further measured by MTT.SKOV3/DDP cells were distributed into the group A and group B.Cells in the group A were added with DDP,group B with DDP and 5μmol/L MIF.The IC50 of SKOV3/DDP cells was measured by MTT at 24,48,and 72 h,respectively,and then we calculated the reversal fold(RF).The cell cycle of SKOV3/DDP was detected by flow cytometry(FCM).Results MTT assay results showed that mifepristone inhibited the proliferation of SKOV3 and SKOV3/DDP cells in a dose-dependent manner.The reversal dose of MIF on SKOV3/DDP was 5μmol/L.The IC50 of SKOV3 and SKOV3/DDP to DDP were3.22 and 14.83μg/ml,respectively,and the RI of SKOV3/DDP to DDP was 4.61.The IC50 of group A was(23.13±0.83),(14.83±0.52),(10.07±0.56)μg/mL at 24,48 and 72 h after culture,and(17.06±0.53),(7.77±0.47)and(4.06±0.69)μg/mL in the group B,respectively,and the RF of MIF to SKOV3/DDP were 1.36,1.91,and 2.48,respectively.After 24,48,and 72 hours of culture,the proportion of SKOV3/DDP cells added with 5μmol/L MIF in the G0/G1 phase increased gradually(48.7%,53.9%,55.1%,and 59.0%),while the proportion of cells in the S phase decreased gradually(13.5%,9.6%,7.7%,and 5.8%).Conclusion MIF(5μmol/L)could inhibit the proliferation of SKOV3/DDP cells and reduce the resistance of SKOV3/DDP cells to DDP,and the possible mechanism might be that MIF can block the cell cycle of SKOV3/DDP in the G0/G1 phase.
