摘  要:目的观察腹腔注射CpG寡核苷酸(CpG oligonucleotides,CpGODN)的急性支气管哮喘(简称哮喘)小鼠支气管肺泡灌洗液(BALF)、血清胸腺基质淋巴细胞生成素(TSLP)表达变化。方法36只SPF雄性BALB/c小鼠随机分为A、B、C、D组,每组各9只。A、B、C组用0.1%卵清百蛋白(OVA)/AL(OH)3混合凝胶0.1 mL于第1、14天腹腔注射致敏,第25~31天1%OVA雾化吸入激发小鼠,每日1次,每次30 min,连续7 d,制备急性哮喘模型。B组激发前1 d和每次激发前1 h,0.5 mg/kg的地塞米松腹腔注射;C组激发前1 d和每次激发前1 h,50μg的CpGODN腹腔注射;D组为正常对照组,每次致敏和激发均以生理盐水代替。末次激发24 h时取各组小鼠,采集眼球血后处死小鼠,收集右肺BALF,取肺组织观察肺组织炎性变化,ELISA法检测BALF、血清中TSLP。结果A组小鼠肺组织炎症水平、超微结构改变明显,B、C组肺组织炎症水平、超微结构改变轻于A组,D组小鼠肺组织无炎症水平改变。A、B、C、D组BALF中TSLP水平分别为(2.481±0.152)、(2.263±0.122)、(2.483±0.191)、(2.483±0.280)pg/mL,与A、C、D组比较,B组小鼠BALF中TSLP水平低(P均<0.05)。A、B、C、D组血清TSLP水平分别为(2.739±0.127)、(3.028±0.142)、(3.556±0.102)、(2.560±0.096)pg/mL,与D组比较,A、B、C组血清TSLP水平升高(P均<0.05),A、B、C组间两两比较,P均<0.05。结论腹腔注射CpGODN可减轻急性哮喘小鼠肺组织炎症水平,其机制可能与CpGODN影响TLSP的生物学效应有关。Objective To observe the expression of bronchoalveolar lavage fluid(BALF)and serum thymic stromal lymphopoietin(TSLP)in asthmatic murine models with intraperitoneal injection of CpG oligonucleotides(CpGODN).Methods Thirty-six SPF male BALB/c mice were randomly divided into groups A,B,C and D,with 9 mice in each group.According to the literature,the acute asthma mouse models were induced in the groups A,B and C.The acute asthma model was sensitized by intraperitoneal injection of 0.1%OVA/AL(OH)3 mixed gel(0.1 mL)on the 1st and 14th days,followed by atomization inhalation of 1%OVA from 25th to 31st days,once a day for 30 min each time,for 7 consecutive days.In the group B,0.5 mg/kg dexamethasone was intraperitoneally injected at 1 day and 1 hour before eachstimulation.In the group C,50μg CpGODN was intraperitoneally injected at 1 day and 1 hour before each stimulation.Group D was the normal control group,and normal saline was used for each sensitization and stimulation.Group A was asthma model group.After the last stimulation for 24 h,eyeball blood of each mouse was collected and then the mice were sacrificed;the right lung BALF was collected,lung tissue was taken to observe the changes of inflammatory,and the TSLP expression in BALF was detected by ELISA.Results The inflammation and ultrastructure of the lung tissues of group A significantly changed as compared with those of the groups B and C,group D did not change.The expression of TSLP in the BALF of groups A,B,C and D were(2.481±0.152),(2.263±0.122),(2.483±0.191),and(2.483±0.280)pg/mL,respectively.Compared with groups A,C and D,the level of TSLP in BALF of mice in the group B was lower(all P<0.05).The expression levels of TSLP in serum of groups A,B,C and D were(2.739±0.127),(3.028±0.142),(3.556±0.102)and(2.560±0.096)pg/mL,respectively.Compared with the group D,the expression of TSLP in serum of groups A,B and C increased gradually(all P<0.05),and the pairwise comparison between groups A,B and C was statistically significant(all P<0.05).Conclusion Int
