摘  要:为建立国内大豆实际生产上可直接利用的大豆品种的大豆体细胞胚再生途径,促进大豆的分化发育、遗传转化、诱变等方面的研究和应用,本研究以大豆主栽品种黑农84为材料,针对影响体细胞胚诱导率的3个重要因素pH、外植体幼胚的直径和激素浓度设计正交试验。结果表明:外植体最适幼胚直径为4~5 mm,诱导培养基最佳配比为2,4-D浓度40 mg·L^-1、蔗糖浓度0.5%和琼脂浓度0.6%,pH7.0,此条件下体细胞胚的发生率为88.6%。同时利用获得的体细胞胚通过增殖培养、分化培养、生根培养和移栽,最终得到了再生植株,再生率为10.2%。The establishment of somatic embryogenesis pathway for main soybean varieties is very important in the study of plant differentiation,genetic transformation and mutation.In this study,three important factors affecting the induction rate of somatic embryos,pH value,the size of immature embryos of explants and hormone concentration,were studied using the main soybean cultivar Heinong 84 as the test material.The results showed that the the size of immature embryos of explants was 4-5 mm.The optimal condition of induction medium was consist of 40 mg·L^-12,4-D,0.5%sucrose,0.6%agar pH7.0.And the incidence of somatic embryogenesis was 88.6%.In this study,the regenerated plants were obtained through proliferation culture,differentiation culture,rooting culture and transplanting,and the regeneration frequency is 10.2%.
