摘 要:目的:优化微波辅助提取白藜芦醇的提取工艺;方法:以提取率为考察目标,以粒度、液料比、提取时间、提取溶剂乙醇浓度为考察单因素,采用中心复合设计法优化提取工艺条件,通过方差分析及线性、二项式回归方程模型拟合,进行预测分析;结果:经优化得到最佳实验条件为药材粒度80目,液料比42:1,微波温度60℃,微波时间6.5 min,微波功率为500 W,76%乙醇做提取溶剂。在此条件下,白藜芦醇得率为3.96 mg/g(RSD=1.53%,n=3),与模型预测值相符。结论:该法优化得到的工艺条件操作简便、提取率及灵敏度更高相较于传统溶剂回流法更高、更准确,为今后白藜芦醇提取及剂型研究奠定基础。Objective:To optimize extraction technology of resveratrol by Microwave-assisted extraction(MAE).Method:Using yield as dependent parameters,Using particle size、ratio of liquid to solid、microwave time、extraction solvent concentration as independent variables,Central composite design-response surface methodology were used to optimize the extracting process by Microwave-Assisted Extraction(MAE),Central composite design and response surface methodology were used to optimize the extracting process.Through variance analysis and linear and binomial regression equation model fitting,the prediction analysis was carried out by comparing observed and predicted values.Results:The optimum extraction conditions were as follows:particle size:80 mesh,ratio of liquid to solid:42:1,microwave temperature:60℃,microwave time:6.5 min,microwave power:500 W,solvent:76%ethanol.the yield of resveratrol can reach3.96mg/g(RSD 1.53%,n=3),which was close to the model predictions.Conclusion:Compared with traditional solvent reflux method(SRE),this method is more simple with higher extraction rate and sensitivity,and provides a reference for the research of resveratrol extraction and dosage form in the future.
