摘 要:目的研究“内消软脉汤”对ApoE^-/-小鼠血脂及高敏C反应蛋白的影响,并探究其是否与解耦联蛋白1依赖性脂解有关。方法将48只ApoE^-/-小鼠随机分为空白组、模型组、“内消软脉汤”低剂量组、“内消软脉汤”高剂量组,每组12只。采用高脂饲养的方法复制高脂血症小鼠模型,造模后各组予相应药物灌胃,连续给药60天后处死小鼠,并取血清用于生化法检测血脂甘油三酯、低密度脂蛋白及高敏C反应蛋白,用HE染色法及Western blot法检测肩胛区棕色脂肪组织、腹股沟皮下白色脂肪组织、附睾内脏白色脂肪组织等脂肪组织中解耦联蛋白1表达。结果与空白组相比,模型组的甘油三酯、高敏C反应蛋白略有降低,但无统计学意义;与模型组相比,“内消软脉汤”低剂量组、高剂量组的甘油三酯、高敏C反应蛋白均下降(P<0.05),解耦联蛋白1的表达随药物呈剂量依赖性升高(P<0.05)。结论“内消软脉汤”可降低ApoE^-/-小鼠的炎症及血脂水平,此作用的实现可能与激活解耦联蛋白1依赖性脂解有关。Objective To study the effects of‘Neixiao Ruanmai decoction’on blood lipid and HS-CRP in ApoE^-/-mice,and to explore whether it is related to UCP1 dependent lipolysis.Methods 48 ApoE^-/-mice were randomly divided into blank group,model group,low dose of‘Neixiao Ruanmai decoction’group and high dose of‘Neixiao Ruanmai decoction’group,12mice in each group.The hyperlipidemia mouse model was established by high-fat feeding method.After the modeling,each group was given corresponding drugs,and the mice were sacrificed after 60 days of continuous administration,and serum was taken for biochemical determination of triglyceride(TG),total cholesterol(TC),low density lipoprotein(LDL)and high sensitivity C-reactive protein(HS-CRP).UCP1’s expression in brown adipose tissues,such as in scapular area(iBAT),subcutaneous white adipose tissue of groin(sWAT)and epididymal visceral white adipose tissue(eWAT),were detected by HE staining and Western bolt.Results Compared with the blank group,the TG and HS-CRP in the model group were slightly decreased,but there was no statistical significance.Compared with the model group,TG and HS-CRP in the low and high dose of‘Neixiao Ruanmai decoction’groups were decreased(P<0.05),and the expression of UCP1 increased in a dose-dependent manner(P<0.05).Conclusion Neixiao Ruanmai decoction can reduce inflammation and lipid levels in ApoE^-/-mice,which may be related to activation of UCP1 dependent lipolysis.
