摘 要:目的利用网络药理学探究由黄芩、黄连、黄柏、栀子组成的黄连解毒汤治疗动脉粥样硬化药效成分的可能靶点。方法根据类药性及口服生物利用度,通过中药系统药理学数据库分析平台、GeneCards和OMIM数据库、反向分子对接服务器,筛选黄连解毒汤治疗动脉粥样硬化的有效活性成分预测作用靶标;采用Cytoscape 3.7.1绘制出黄连解毒汤活性成分-动脉粥样硬化作用靶标网络图形;应用STRING数据库制作靶标相互作用网络并运用Network Analyzer作网络分析;最终在生物学信息注释数据库中进行基因本体生物过程分析,同时在京都基因与基因组百科全书通路富集分析。结果本研究共预测得到筛选出102个主要活性成分,其中黄芩36个,黄连14个,黄柏37个,栀子15个。得到活性成分的作用靶标170个,包括AKT1、EGF、JUN、MMP9、FOS等;GO生物过程结果显示与蛋白质异二聚化活性、辅因子结合、内肽酶活性等相关,KEGG通路富集分析结果显示主要涉及MAPK信号通路、磷脂酰肌醇3激酶(PI3K)信号通路、VEGF信号通路、NF-κB信号通路等方面。结论本研究体现了中药复方黄连解毒汤多成分、多靶点及多途径的作用特点,为黄连解毒汤干预动脉粥样硬化作用机制提供新思路和新靶点。Objective Network pharmacology was used to explore the possible target of Huanglian Jiedu Decoction composed of Huangqin(Radix Scutellariae), Huanglian(Rhizoma Coptidis), Huangbai(Cortex Phellodendri) and Zhizi(Fructus Gardeniae) in the treatment of atherosclerosis.Methods According to the medicinal properties and oral bioavailability, the prediction targets of effective active components of Huanglian Jiedu Decoction in the treatment of atherosclerosis were screened through the pharmacology database analysis platform of traditional Chinese medicine system, GeneCards and OMIM database and reverse molecular docking server. The target network graph of active component of Huanglian Jiedu Decoction was drawn by Cytoscape 3.7.1, and the target interaction network was made by STRING database and analyzed by Network Analyzer. Finally, the gene ontology biological process analysis was carried out in the biological information annotation database, and the gene and genome encyclopedia pathway was enriched and analyzed at the same time. Results In this study, 102 main active components were screened, including 36 from Huangqin(Radix Scutellariae), 14 from Huanglian(Rhizoma Coptidis), 37 from Huangbai(Cortex Phellodendri) and 15 from Zhizi(Fructus Gardeniae). A total of 170 active targets were obtained, including AKT1, EGF, JUN, MMP9, FOS and so on. The results of GO biological process showed that it was related to protein heterodimerization activity, cofactor binding, endopeptidase activity and so on. The results of KEGG pathway enrichment analysis showed that it was mainly related to MAPK signal pathway, phosphatidylinositol 3 kinase(PI3 K) signal pathway, VEGF signal pathway, NF-κB signal pathway and so on. Conclusions This study reflects the characteristics of multi-components, multi-targets and multi-ways of Huanglian Jiedu Decoction, and provides new ideas and new targets for the mechanism of Huanglian Jiedu Decoction in the intervention of atherosclerosis.
