当归四逆汤加减调节Akt/mTOR信号诱导自噬治疗子宫内膜异位症的研究

摘　　要：目的探讨当归四逆汤加减中药治疗子宫内膜异位症作用机理。方法将75只模型大鼠分为5组,加假手术正常组,各15只,分别予当归四逆汤加减中药高、中、低剂量、内美通胶囊、生理盐水灌胃给药。结果给药后中高组及西药组卵巢异位病灶缩小(P<0.05)。中高组、西药组在位、异位内膜AKT、mTOR表达水平低于模型组(P<0.05),LC-3及Beclin-1表达水平及血清TIMP-2水平高于模型组(P<0.01),中高、中中、中低组、孕三烯酮组血MMP-9、VEGF、SICAM-1水平明显低于模型组(P<0.05)。结论当归四逆汤加减中药可有效缩小异位灶,抑制Akt/mTOR通路,提高自噬基因Beclin-1、LC3活性,降调VEGF、SICAM-1水平,调整MMPs/TIMPs的表达比例,抑制异位内膜生长。Objective To explore the mechanism of treating endometriosis with traditional Chinese medicine by Danggui Sini Decoction. Methods Seventy-five successfully modeling endometriosis rats were randomly divided into 5 groups:high-dose,middle-dose and low-dose groups of Danggui Sini Decoction,nemestran group and blank control, respectively. The sham surgery rats were treated as normal control group. The three dose groups were treated with different concentrations of Danggui Sini Decoction. Nemestran was administrated to the nemestran group and 0.9% NaCl to blank and normal groups,respectively. Results The volume of the heterotopic focus in high-dose group was decreased obviously after administration(P<0.05). Protein and mRNA expressions of AKT and mTOR in high-dose and nemestran groups were significantly lower than that in the model group, but protein and mRNA expressions of Beclin-1 and LC3 were significantly higher than that in the model group(P<0.05). The serum levels of TIMP-2 was increased too. Compared with those of the model group, serum levels of VEGF, SICAM-1 and MMP-9 in low-dose, middle-dose, high-dose and nemestran groups were decreased significantly(P<0.05). Conclusion Danggui Sini Decoction can reduce the ectopic focus effectively, inhibit Akt/mTOR pathway and adjust the expressions of MMP-9, TIMP-2, VEGF, SICAM-1, Beclin-1 and LC3. Then it can inhibit the growth of ectopic endometrium.
