摘 要:目的探讨人参皂苷3(Rg3)通过缝隙连接细胞间通讯(GJIC)发挥生精保护作用。方法将40只初成年SD雄性大鼠随机分为对照组、Rg3组(20 mg/kg)、DBP组(500 mg/kg)及(Rg3:20 mg/kg+DBP:500 mg/kg)组。连续给药35 d后,处死各组大鼠,检测精子活力、浓度、畸形率;放射免疫法检测血清睾酮(T)、黄体生成素(LH)、卵泡刺激素(FSH)水平;流式细胞仪检测睾丸组织细胞周期,光镜观察睾丸组织形态结构,Western Blot检测睾丸组织Cx43蛋白。结果与对照组比较,DBP模型组睾丸组织有明显的组织结构损伤,精子密度和精子活力下降,血清黄体生成素(LH)、睾酮(T)下降,卵泡刺激素(FSH)上升,组织细胞周期G0/G1期细胞比例增多,Cx43蛋白表达量降低,差异有统计学意义(P<0.05);与DBP组比较,DBP+Rg3组睾丸组织结构趋于正常,精子密度和精子活力上升,睾酮(T)和黄体生成素(LH)水平上升,卵泡刺激素(FSH)下降,G0/G1期细胞比例降低,Cx43的表达量上升,差异有统计学意义(P<0.05)。结论 Rg3可通过GJIC发挥生精保护作用,改善生精障碍,具有生精保护作用。Objective To study the effect of ginseng saponin 3(Rg3) on protecting spermatogenesis in rats by GJIC. Methods Forty adult male SD rats were randomly divided into control group, Rg3 group(20 mg/kg), DBP group(500 mg/kg) and Rg3+DBP group. After continuous administration for 35 days, rats in each group were sacrificed, and the sperm motility, concentration and abnormal rate were measured. The serum levels of T,LH and FSH were measured by radioimmunoassay. Flow cytometry was used to detect the cell cycle of testis tissue. The morphology and structure of testis tissue were observed under light microscope. Cx43 protein was detected by Western Blot. Results Compared with those of the control group, the testicular tissue of the DBP model group had obvious tissue damage. The sperm density and sperm motility decreased. The serum levels of LH and T decreased, FSH increased and G0/G1 cells percentage increased. The expression of Cx43 protein decreased, and the difference was statistically significant(P<0.05). Compared with those of the DBP group, the testicular tissue structure in the DBP+Rg3 group tended to be normal. The sperm density and sperm motility increased, levels of T and LH increased, FSH decreased and G0/G1 cells decreased, the expression of Cx43 increased, and the difference was significant(P<0.05). Conclusion Rg3 exerts spermatogenesis protection and improves spermatogenesis disorder through GJIC.
