摘 要:目的探讨不同浓度地西他滨与CD4^+T淋巴细胞的结合。方法正常人外周血的单个核细胞(PBMCs)与不同浓度(0、0.2、0.4、0.8μm和1.6μm)N末端标记FITC荧光的地西他滨37℃共孵育1h,流式细胞仪初步分析地西他滨与人PBMCs中CD4^+T淋巴细胞的结合,以及两者结合的最适浓度,实验重复8次;利用美天旎磁珠分选CD4^+T淋巴细胞,与最适浓度N末端标记FITC荧光的地西他滨37℃共孵育1 h,流式细胞仪分析地西他滨与分选CD4^+T淋巴细胞的结合率,实验重复8次。结果不同浓度地西他滨可以和人PBMCs中的CD4^+T淋巴细胞结合,其平均结合率为2.05%±0.09%,95%置信区间为1.02%~1.43%,且当其浓度为0.8μm时,两者的结合最明显,即该浓度为两者结合的最适浓度。且最适浓度地西他滨可以与美天旎磁珠分选CD4^+T淋巴细胞结合,平均结合率16.64%±1.62%,95%置信区间为14.06%~21.35%。结论地西他滨可以与人PBMCs中及分选CD4^+T淋巴细胞结合,并可能对其免疫作用的发挥产生影响。Objective To investigate the combination of different concentrations of decitabine and CD4^+T lymphocytes.Methods We first determined the binding efficiency of CD4^+T celsl in human peripheral blood mononuclear cells(PBMCs)to different concentrations(0μm,0.2μm,0.4μm,0.8μm and 1.6μm)of N-terminal FITC-labeled decitabine by flow cytometry after that they were incubated at 37℃for 1 hour.The experiment was repeated for 8 times.The CD4^+T lymphocytes were separated by Meitianni magnetic beads and incubated with the optimal concentration of N-terminal FITC-labeled decitabine at 37℃for 1 hour.The binding rate between dacitabine and sorted CD4^+T lymphocytes was analyzed by flow cytometry.The experiment was repeated for 8 times.Results Different concentrations of decitabine could bind to CD4^+T lymphocytes in human PBMCs.The average binding rate was(2.05%±0.09%),and the 95%confidence interval was 1.02%~1.43%.When the concentration was 0.8μm,the combination of the two is the most obvious,that is,the concentration is the optimum concentration for the combination of the two.Decitabine of the optimum concentration of could bind to CD4^+T lymphocytes,the average binding rate was(16.64%±1.62%),and the 95%confidence interval was 14.06%~21.35%.Conclusion Decitabine can bind to CD4^+T lymphocytes,and may have an effect on its immune function.
