摘  要:目的:探讨阿魏酸钠(SF)对缺氧缺血诱导PC12细胞损伤的保护作用及其机制。方法:将PC12细胞分为4组,分别是对照组(control)、缺血缺氧模型组(HI)、阿魏酸钠处理组(HI+SF)和LNA-miR-9干预组(HI+LNA),以低血清-氧剥夺的方法建立PC12细胞缺血缺氧(HI)模型,经阿魏酸钠和microRNA-9(miRNA-9)抑制剂LNA-miR-9处理后,利用real time RT-PCR和Western Blot检测各组PC12细胞中miRNA-9、乙酰胆碱酯酶(ACh E)和脑源性神经生长因子(BDNF)的mRNA及蛋白表达水平。结果:低血清-氧剥夺能明显诱导PC12细胞的损伤,与control组相比,HI组PC12细胞变圆,贴壁减少,悬浮的死细胞增多,且细胞内的miRNA-9、ACh E和BDNF表达明显增加(P <0.05);与HI组相比,HI+SF组PC12细胞贴壁明显增加,细胞突起增多,悬浮的死细胞明显减少,且细胞中的miRNA-9表达明显降低(P <0.05);HI+LNA组PC12细胞较HI组贴壁增加,细胞突起增多,悬浮的死细胞明显减少,且细胞中的ACh E和BDNF表达明显降低(P <0.05);同时,与HI组相比,HI+SF组中的ACh E和BDNF表达明显降低(P <0.05)。结论:SF对PC12细胞缺血缺氧损伤具有神经保护作用,其机制可能是通过SF抑制miRNA-9,从而降低ACh E和BDNF的表达有关。Objective: To investigate the protective effect and mechanism of sodium ferulic( SF) on PC12 cells injury induced by hypoxia-ischamia. Methods: The PC12 cells were divided into four groups: control group( control),ischemia hypoxia model group( HI),sodium ferulic treatment group( HI + SF),and LNA-miR-9 intervention group( HI + LNA),PC12 cell model of starve serum-oxygen deprivation has been established to mimic cerebral hypoxia-ischemia injury. After treatment with SF and miRNA-9 inhibitor LNA-miR-9,real time RT-PCR and Western Blot were used to detect miRNA-9,acetyl cholinesterase( ACh E),and brain-derived neuotrophyic factor( BDNF) mRNA level as well as protein level in cells,respectively. Results: The starve serum-oxygen deprivation can obviously induced PC12 cells damage. Compared with the control group,PC12 cells in the HI group were rounded,adherent decreased,suspended dead cells increased,and the expression of miRNA-9,ACh E,and BDNF in the cells was significantly increased( P <0. 05). Compared with the HI group,the adhesion of PC12 cells in the HI + SF group was significantly increased,the number of cell protrusions was increased,the suspended dead cells were significantly reduced,and the expression of miRNA-9 in the cells was significantly reduced( P < 0. 05). Compared with the HI group,PC12 cells in the HI + LNA group had increased adherence,increased cell protrusion,and significantly reduced suspended dead cells,and the expressions of ACh E and BDNF in the cells were significantly lower than those in HI group( P < 0. 05). At the same time,compared with the HI group,the expressions of ACh E and BDNF in the HI + SF group were significantly reduced( P < 0. 05). Conclusion: The SF has neuroprotective effect on hypoxic-ischamic injury of PC12 cells. The mechanism may be that SF inhibits the expression of miRNA-9,thereby reducing the expressions of ACh E and BDNF.
