摘  要:目的:探讨消癌解毒方防治结直肠腺瘤性息肉癌变的潜在机制。方法:运用中药系统药理学数据库收集消癌解毒方所含化合物对应的相关靶点;使用OMIM、TTD、PharmGkb、DiGSeE数据库,分别检索与肠腺瘤(Colorectal Adenoma)、肠癌(Colorectal cancer)相关的疾病靶点;使用Cytoscape 3.2.1软件进行"消癌解毒方-化合物-关键靶点"网络关联,并通过Enrichr数据库,进行关键靶点的基因本体(gene ontology,GO)和基因组百科全书(kyoto encyclopedia of genes and genomes,KEGG)富集分析。结果:消癌解毒方所含化合物对应靶点243个、肠腺瘤对应靶点3279个、肠癌对应靶点4119个,三者交集的关键靶点110个。关键靶点的GO富集条目有2522条,其中生物过程相关的条目2033条,细胞组成相关的条目132条,分子功能相关的条目357条;关键靶点的KEGG富集通路涉及PI3K-Akt信号通路、IL-17信号通路、肿瘤坏死因子信号通路、细胞衰老、细胞凋亡等。结论:消癌解毒方防治结直肠腺瘤性息肉癌变的潜在作用机制可能与调控免疫炎症反应、细胞增殖及凋亡、机体衰老及体内激酶活性等过程有关,具有多靶点、多途径、多系统的机制特点。Objective:To observe the potential mechanism of Xiaoai Jiedu Prescription in preventing and treating colorectal adenoma canceration.Methods:The pharmacology database of the Chinese medicine system was used to collect the relevant targets corresponding to the compounds contained in the Xiaoai Jiedu Prescription;OMIM,TTD,PharmGkb,and DiGSeE databases were used to search for disease targets related to colorectal adenoma canceration and colorectal cancer,respectively;Cytoscape 3.2.1 software was used to conduct network association of"Xiaoai Jiedu Prescription-compounds-key target",and GO(Gene Ontology)and KEGG(Signal Pathway)enrichment analysis of key target was carried out through Enrichr database.Results:There are 243 compounds,3279 colorectal adenoma canceration and 4119 colorectal cancer targets in Xiaoai Jiedu Prescription,and 110 key targets intersect among them.There are 2522 items of GO enrichment,including 2033 items related to biological process,132 items related to cell composition and 357 items related to molecular function;key target KEGG enrichment pathway involves PI3 K-Akt signaling pathway,IL-17 signaling pathway,tumor necrosis factor signaling pathway,cell aging,apoptosis and other pathways.Conclusion:The potential mechanism of Xiaoai Jiedu Prescription in the prevention and treatment of colorectal adenoma canceration may be related to the regulation of immune inflammatory reaction,cell proliferation and apoptosis,body aging and in vivo kinase activity.It has the characteristics of multi-target,multi-channel and multi-system mechanism.
