摘 要:目的观察温胆汤对脑缺血/再灌注损伤模型大鼠行为学、海马中5-羟色胺(5-HT)、丙二醛(MDA)、多巴胺(DA)、β-内啡肽(β-EP)表达的影响。方法将30只大鼠随机分为空白组、模型组、尼莫地平组、温胆汤高剂量组、温胆汤低剂量组5组,每组各6只,空白组仅分离颈内动脉,给予生理盐水10 mL/kg灌胃;其余大鼠采用Longa法线栓建立缺血/再灌注模型,模型组给予生理盐水10 mL/kg灌胃;尼莫地平组给予尼莫地平悬浮液20 mg/kg;温胆汤高剂量组和低剂量组分别给予温胆汤4 g/kg、2 g/kg,给药体积及给药频率均相同。连续给药14 d后对所有大鼠进行Morris水迷宫测试。完成行为学检测后,分离出大鼠双侧海马,采用酶联免疫吸附法(ELISA)测定海马组织中DA、5-HT、β-EP、MDA的表达。结果与空白组比较,各组动物水迷宫游泳总路线均明显增加,与模型组比较,各给药组水迷宫游泳总路线明显缩短,其中温胆汤高剂量组在D3、D5时差异显著(P<0.05),在D4时有差异极显著(P<0.01);温胆汤低剂量组在D4时差异显著(P<0.05)。5-HT各给药组较模型组有下降趋势,但差异无统计学意义(P>0.05);DA各给药组较模型组有上升趋势,差异有统计学意义(P<0.05);β-EP各给药组较模型组有下降趋势,且温胆汤高剂量组差异显著(P<0.05);MDA各给药组较模型组均有下降趋势,差异有统计学意义(P<0.05)。结论缺血/再灌注可导致海马神经元损伤,温胆汤能有效改善脑缺血/再灌注大鼠的神经损伤症状,影响海马组织中细胞因子及神经递质的含量。Objective To observe the effect of Wendan Decoction on the ethology of rats with cerebral ischemia/reperfusion injury and the expressions of 5-hydroxytryptamine(5-HT),malonaldehyde(MDA),dopamine(DA)andβ-endorphin(β-EP)in their hippocampi.Methods Thirty rats were randomly divided into five groups:blank group,model group,Nimodipine group,Wendan Decoction high-dose group and Wendan Decoction low-dose group,with 6 rats in each group.In the blank group,only internal carotid artery separation was performed and normal saline(10 mL/kg)was given to the rats by gavage.The rats in other groups were treated with Longa to establish the ischemia/reperfusion model.The rats in the model group were given normal saline(10 mL/kg)by gavage.Rats in Nimodipine group were given Nimodipine suspension liquid by 20 mg/kg.Rats in the Wendan Decoction high-dose and low-dose groups were given Wendan Decoction by 4 g/kg and 2 g/kg respectively.All the dosage volume and frequency were the same.All rats were given Morris water maze tests after 14 days of continuous administration.After completing the behavioral test,the rats'bilateral hippocampi were isolated,and the expressions of DA,5-HT,β-EP and MDA in the hippocampus were determined by ELISA.Results Compared with the blank group,the total swimming routes of the other groups were significantly increased.Compared with the model group,the total swimming routes of drug administration groups were significantly shortened.There were significant differences between the Wendan Decoction high-dose group and the model group at D3 and D5(P<0.05),and high statistically difference at D4(P<0.01).There was significant difference between the model group and the Wendan Decoction low-dose group at D4(P<0.05).Compared with the model group,5-HT expression in each drug administration group showed a downward trend,but there was no significant difference(P>0.05).Compared with the model group,DA expression in each drug administration group showed an upward trend,and there were significant differences between tho
