海洛因对仔鼠肝组织结构和抗氧化物酶活性的影响

摘　　要：为了探讨海洛因对仔鼠肝组织结构和抗氧化物酶活性的影响,选取60只昆明小鼠随机分为海洛因组和对照组组,用递增剂量(2,3,4 g·L^-1)给海洛因组连续(1~5,6~10和11~15 d)腹腔注射海洛因溶液15 d(每次0.2 mL,每天2次),给对照组注射等量生理盐水,用比色法检测仔鼠血浆谷草转氨酶(GOT)、谷丙转氨酶(GPT)活性和肝组织硫氧还蛋白还原酶(TrxR)、髓过氧化物酶(MPO)活性及丙二醛(MDA)含量的变化,用生物显微技术观察仔鼠肝组织结构的变化.结果显示,与对照组比较,海洛因组仔鼠血浆GOT,GPT活性均显著升高(P<0.01),而肝组织TrxR活性显著降低(P<0.01),肝组织MPO活性则显著升高(P<0.05),肝组织MDA含量明显升高(P<0.01),肝组织细胞出现不同程度的损伤.表明海洛因影响细胞抗氧化酶活性,对肝组织结构有明显的损伤作用.To explore the effect of heroin on the liver tissue structure and antioxidant enzyme in filial mice,sixty mice were randomly divided into 2 groups,such as the control group and the heroin group.The mice of heroin group were injected intraperitoneally with heroin of the increasing doses(2,3,4 g·L^-1)continuous(1~5,6~10 and 11~15 d)for 15 d(twice a day),and the control group was injected with equivalence saline.Activities of glutamic-oxalacetic transaminase(GOT),glutamic-pyruvic transaminase(GPT)in blood,and thioredoxin reductas(TrxR),myeloperoxidase(MPO)in hepatic tissue,as well as content of malonaldehyde(MDA)were detected by colorimetry,and the morphological changes of hepatic tissues were observed under microscopy.The results show that compared with the control group,the activities of plasma GPT and GOT were significantly increased in the herion group(P<0.01),and the activity of hepatic tissue TrxR decreased obviously(P<0.01),while the activity of MPO and the contents of MDA were increased significantly in the herion group(P<0.05 or P<0.01).The hepatic tissues structure appear different degrees damage in the herion group.All results show that heroin affects the activity of cell antioxidant enzymes and heroin has obvious damage effect on liver tissue structure.
